摘要
核因子κB(NF-κB)普遍存在于真核生物的细胞中,是重要的诱导性转录因子,在细胞的生长、增殖、分化、凋亡,以及癌变过程中均发挥重要作用。当细胞受到各种内外界的刺激,NF-κB信号通路被激活,核因子与相应基因结合从而调节靶基因的表达。研究发现NF-κB信号通路激活主要有经典途径和旁路途径两种。同时还发现,NF-κB信号通路的激活和抑制会相应的调节细胞各种生理生化反应。在机体中,NF-κB信号通路的激活往往伴有肿瘤或炎症的发生。文章主要综述了NF-κB信号通路相关蛋白的结构和功能,以及NF-κB信号通路的激活和抑制在治疗肿瘤和炎症等疾病方面的的最新研究进展。对NF-κB信号通路相关靶点的研究,为新的消炎和抗肿瘤药设计以及疾病的治疗等提供有益的思路。
Nuclear factor kappa B( NF-κB) is an induced transcription factor which is commonly found in eukaryotes. It plays an important role in all stages of cell cycle,including cell growth,proliferation,differentiation,apoptosis,cancer,etc. When the cell is stimulated by various internal and external stimuli,NF-κB signaling pathway will be activated. The nuclear factor in the cytoplasm will enter into the nucleus. Accordingly it is combined with the corresponding gene and then regulated the transcription and expression of target genes. In the last few years,with the development of the deep-going research of NF-κB and NF-κB signal pathway,more and more studies have found that there are two mainly different ways in NF-κB pathway activation. It was generally called classical pathway and the alternative pathway. At the same time,many studies have found that activating or inhibiting NF-κB signaling pathway can regulate the physiological and biochemical responses of cells. For example,as activation of NF-κB was associated with the occurrence of many malignant tumors and metastasis. Because NF-κB will promote the gene in some tumor cells high expression a number of immunosuppressive agents and anti-inflammatory drugs are now used to play a role in preventing kappa B inhibitory factor degradation or preventing the NF-κB binding to the kappa B site of the target gene. The structure and function of the NF-κB signaling pathway related proteins as well as the newest research progress in the activation or inhibition utility of NF-κB signaling pathway for the treatment of inflammatory and tumor or other diseases were reviewed in this paper. This article is expected to provide useful ideas for the new antiinflammatory and anti tumor drug design and the treatment of related diseases through the review of the NF-κB signaling pathway.
