摘要
目的探讨大黄素对兔主动脉平滑肌细胞(CCC-SMC-1)凋亡基因Fas mRNA表达的影响。方法 MTT法观察大黄素对兔主动脉平滑肌细胞增殖的抑制作用,RT-PCR法检测大黄素对兔主动脉平滑肌细胞中Fas mRNA的表达。结果MTT法显示大黄素在0.05~0.25 mmol·L^(-1)浓度范围内对CCC-SMC-1细胞增殖有抑制作用,作用24 h后的IC_(50)为0.10 mmol·L^(-1),且IC_(50)随作用时间的延长显著前移。RT-PCR法显示,不同剂量的大黄素作用于兔主动脉平滑肌细胞24 h后,与正常组细胞相比,Fas mRNA的表达水平均升高(P<0.05)。结论大黄素能抑制CCC-SMC-1细胞的增殖,其机制可能与上调的Fas mRNA表达有关。
Objective The purpose of this study is to research the inhibition of emodin on rabbit aortic smooth muscle cell proliferation( CCC- SMC- 1) and the effect on apoptosis gene of Fas expression levels. Methods The rabbit aortic smooth muscle cells was cultured,to observe emodin on rabbit aortic smooth muscle cell proliferation by MTT method. The emodin impact on gene expression of relative amount of Fas of rabbit aortic smooth muscle cells were found with the help of PCR. Results MTT was suggested that emodin have the inhibition effect on rabbit aortic smooth muscle cell proliferation when the concentration in the range of 0. 05 mmol·L^(-1)~ 0. 25 mmol·L^(-1). The 50% inhibitory concentration( IC_(50)) was 0. 1 mmol·L^(-1)after drug intervention for 24 h,and IC_(50) was significantly forward with the extension of action time. The results of RT- PCR showed that the expression of Fas gene was significantly increased( P〈0. 05) after 24 h compared with normal cells through different dose of emodin impact on rabbit aortic smooth muscle cell. Conclusion Emodin can inhibit the proliferation of CCC- SMC- 1cell,its mechanism may be related to rise the expression of Fas mRNA.
