摘要 为探索SCOT标记在油菜遗传多样性研究中的应用,本利用36对有多态性的SCOT标记和在油菜多样性分析中广泛使用的30对SRAP标记,对65份陕南地区甘蓝型油菜高代材料进行遗传多样性分析。结果表明,SCOT标记平均每个标记检测到5个等位变异,每个SCOT位点的遗传多态性信息含量在0.28~0.81,平均值为0.57,供试材料的遗传相似系数集中在0.56~0.89。平均每个SRAP标记检测到4个等位基因,每个SRAP位点的遗传多态性信息含量在0.17~0.85,平均值为0.51。供试材料的遗传相似系数集中在0.50~0.88。本试验得出,SCOT标记的聚类结果和SRAP标记近似,SCOT标记的多态性高于SRAP标记,SCOT标记可以用于油菜遗传多样性的研究。 In order to explore the utilization of start codon targeted polymorphism( SCOT) molecular marker technique in the research of genetic diversity in Brassica napus,thirty six pairs of primers as SCOT markers and 30 pairs of SRAP markers widely used in the analysis of rape variety were utilized to reveal genetic diversity of 65 Brassica napus with advanced inbreeding line materials in southern areas of Shaanxi province. The result showed that on average,5 allelic variations were detected by each SCOT marker,the genetic polymorphism information content of each SCOT locus was between 0. 28 and 0. 81,of which the average was 0. 57 for the tested materials,and the genetic similarity coefficient was between 0. 56 and 0. 89. In addition,4 allelic variations were verified by each SRAP marker in average,genetic polymorphism information content of each SRAP locus was between 0. 17 and 0. 85,the average value was 0. 51,and the genetic similarity coefficient was between 0. 50 and 0. 88 for these tested materials. It was concluded that the clustering results of SCOT markers were similar with results of SRAP markers. By comparison,the polymorphic rate verified by SCOT markers was higher than that by SRAP markers. As an alternative molecular marker,SCOT could be used to study the genetic diversity of Brassica napus.
