摘要
目的分析新疆哈萨克族CETP-Taq IB和ApoA5-1131T/C基因多态性与T2DM关系。方法新疆阿勒泰地区布尔津县哈萨克族252人分成Ⅱ型糖尿病(T2DM)组(126人)、糖耐量异常(IGT)组(63人)和正常糖耐量(NGT)组(63人),应用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)方法对CETP-TaqIB和ApoA5-1131T/C进行基因多态性分析,探讨基因多态性与哈萨克族Ⅱ型糖尿病关联。结果 3组间CETP-TaqIB、ApoA5-1131T/C基因等位基因频率无差异(P>0.05),ApoA5-1131T/C位点基因型频率无差异(P>0.05),CETP-TaqIB位点基因型频率的分布有统计学意义(P<0.05)。CETP-TaqIB的B1B2基因型FPG[(6.24±2.24)mmol/L]明显高于B1B1和B2B2基因型,ApoA5-1131T/C基因TT[(6.09±2.21)mmol/L]基因型的FPG明显高于TC和CC基因型(P<0.05)。结论新疆哈萨克族CETP-TaqIB基因多态性与T2DM相关,哈萨克族ApoA5-1131T/C基因TT基因型FPG水平高于TC和CC基因型。
Objective To study the relationship between gene polymorphism of CETP-Taq IB and ApoA5-1131T/C and type 2 diabetes among Kazaks.Methods A total of 252 Kazaks subjects were selected from Buerjin of Altay prefecturein in Xinjiang,and subsequently divided into groups of type 2 diabetes（T2DM）consisting of 126 subjects,impaired glucose tolerance（IGT）consisting of 63 subjects and normal glucose tolerance（NGT）consisting of 63 subjects.The gene polymorphism of CETP-Taq IB and ApoA5-1131T/C and relationship between gene polymorphism and type 2 diabetes were then analysed using the method of polymerase chain reaction-restriction fragment length polymorphism.Results Among the 3 groups,CETP-TaqIB and ApoA5-1131T/C did not differ significantly in terms of allele and gene frequency（P〈0.05）;the genotypes frequency of ApoA5-1131T/C did not show statistical significance（P〉0.05）;the distribution of genotype frequency of CETP-TaqIB had significant statistical implications（P〈0.05）.FPG（6.24±2.24）mmol/L of genotype B1B2 of CETP-TaqIB was significantly higher than that of genotypes B1B1 and B2B2;FPG of genotypes TT of ApoA5-1131T/C was significantly higher than that of genotypes TC and CC（P〈0.05）.Conclusions CETP-TaqIB gene polymorphism is closely related to T2 DM among Kazaks in Xinjiang;FPG of genotype TT of ApoA5-1131T/C is significantly higher than that of genotypes TC and CC among Kazaks.
