摘要 目的探讨转化生长因子-β1(TGF-β1)对小鼠子宫内膜基质细胞基质金属蛋白酶-9(MMP-9)表达的影响。方法分离和纯化小鼠子宫内膜基质细胞,在此细胞培养体系中加入不同浓度的TGF-β1,终浓度为0.1、2.5、5.0、10.0、20.0ng/mL,培养48h后通过ELISA法检测培养液中MMP-9的含量。结果与对照组比较,实验组随TGF-β1浓度的增加,小鼠子宫内膜基质细胞MMP-9表达上升,差异有统计学意义(P<0.01)。结论小鼠子宫内膜基质细胞体系中,TGF-β1可能通过调控MMP-9的表达,参与调节细胞外基质代谢。 Objective To investigate the effect of transforming growth factor-β1(TGF-β1)on the expression of matrix metalloproteinase-9(MMP-9)in cultured mouse endometrial stromal cells.Methods After separation and purification,the mouse endometrial stromal cells were cultured with different concentrations of TGF-β1,and the final concentrations were 0.1,2.5,5.0,10.0,20.0ng/mL,respectively.ELISA method was used to detect the content of MMP-9in the the culture medium after 48 hculture.Results Compared with the control group,the expression of MMP-9in mouse endometrial stromal cells in the experimental groups increased with the TGF-β1concentration elevation,and the difference was statistically significant(P〈0.01).Conclusion In the mouse endometrial stromal cell system,TGF-β1may be involved in the regulation of extracellular matrix metabolism through regulating the expression of MMP-9.
