摘要
以一株生长性状优良的Cordyceps militaris FFCC5111为出发菌株,通过复合诱变,筛选高产Cordycepin双重营养缺陷型(Xan-+Gua-)菌株。采用200 W、22kHz超声强度联合1%DES复合诱变,获得双重营养缺陷型突变株7株,经抗菌试验、摇瓶培养/发酵复筛至第16天,突变株C.militaris FFCC5111-c产生的Cordycepin总量为4.142g/L,比亲株提高44.5%。试验证明,筛选Xanthine和Guanine双重营养缺陷型突变株是获得Cordycepin高产菌种的有效方法,Cordycepin的生物合成与Guanine代谢相关。
Mutation of Cordyceps militaris FFCC5111 by compound-mutation technique with 1% DES and 200 W,22 kHz ultrasound was performed,and 7 classes of xanthine and guanine double auxotroph mutants were obtained,of which one class was selected as promising preliminary mutant having an antibacterial ability as an index of cordycepin production.Among the double auxotroph mutants,it was shown that mutant C.militaris FFCC5111-c was much better than the others in the cordycepin productivity.The cordycepin production obtained by the C.militaris FFCC5111-c mutant was 4.142g/L in 16-day culture,which was enhanced by 45% more than the original strain.The results indicated that screening Xanthine and Guanine auxotrophic mutant C.militaris was an effective way to obtain a superior production performance of cordycepin and the synthetic pathways of cordycepin and guanine might be linked to each other.
