雌激素受体β过表达对三阴乳腺癌细胞生物学行为的影响 被引量:1

EFFECT OF ERβ'S OVER-EXPRESSION ON TRIPLE- NEGATIVE BREAST CANCER CELL BIOLOGICAL BEHAVIOUR

摘要 目的检测ERβ在三阴性乳腺癌(Triple-Negative Breast Cancer,TNBC)及癌旁组织中的表达,探讨其对TNBC细胞生物学行为的影响,揭示其在TNBC发生发展中的作用。方法免疫组化法检测ERβ在TNBC及癌旁组织中的相对表达量,将ERβ转入低表达的TNBC细胞MDA-MB-435s中,通过MTT试验、Transwell小室侵袭实验及软琼脂集落形成实验,检测MDA-MB-435s生物学行为的变化。结果ERβ在TNBC组织中仍有表达,表达率高达38.33%,在对应的癌旁组织中,表达率为51.67%。ERβ过表达后MDA-MB-435s细胞的增殖和侵袭能力下降,成瘤能力也下降。结论 ERβ在TNBC组织中仍有表达,表达率低于癌旁组织,在TNBC细胞的增殖、侵袭和迁移中起负性调节作用,可作为预测TNBC预后的分子标志物及基因治疗的靶点。 Objective To detect the expression of ERβ in triple - negative breast cancer(TNBC) and para - carcinoma tissue. And to discuss the effect of ERβ on TNBC cell biological behaviour and to reveal its function in the occurrence and development of TNBC. Method The relative expression of ERβ in TNBC and para - carcinoma tissue was detected by IHC, and ERβwas transferred to TNBC cell line MDA -MB -435s where ERβ is under- expressed. MDA -MB -435s' biology behavior changes were detected through the MTr test, transwell cell invasion assay and Colony formation test. Result The expression rate of ERβ was as high as 38.33% in TNBC tissue, and 51.67% in paracarcinoma tissue. The over expression of ERβ inhibited the proliferation, invasion and tumor formation of MDA - MB - 435 s cell. Conclusion ERβ has a lower expression rate in TNBC tissue than in para - carci- noma tissue which could negatively regulate the proliferation, invasion and migration of TNBC and taken as the molecular maker of predicting TNBC prognosis and target of new theranv for TNBC.
