摘要
采用单因素实验研究醇沉时间、乙醇浓度和发酵液pH对香菇(Lentinula edodes)发酵液中提取胞外多糖产量的影响,结果表明最佳醇沉时间为16h,乙醇浓度为80%,pH为8。在单因素实验基础上采用BoxBehnken响应面设计法优化香菇发酵液中胞外多糖提取工艺,最佳提取工艺为:醇沉时间17.71h,乙醇浓度78.66%,pH 7.93。在此条件下,香菇胞外多糖产量达到3.15g/L,与模型预测值3.08g/L接近。
Selected parameters (ethanol precipitation time, ethanol concentration, pH) affecting extracellular polysaccharide (EPS) extraction yields from Lentinula edodes spent culture fluid were optimized by single factorial experiments, and optimal values were determined to be 16 h, 80% and pH 8, respectively. Application of Box-Behnken designs for response surface methodology predicted the optimal extraction conditions to be 17.71 h precipitation time, 78.66% ethanol concentration and pH 7.93. The EPS yield obtained in a verification experiment was 3.15 g/L compared with a predicted yield of 3.08% g/L.
