摘要
目的研究双参强心方(SSQX)对心力衰竭模型大鼠的血流动力学和心肌能量代谢的影响。方法 SD大鼠分为5组,采用左冠状动脉前降支结扎建立心梗后心衰模型,假手术组和模型组给予0.9%生理盐水,双参强心低剂量、中剂量和高剂量组分别给予双参强心方含生药量8.4g/kg、16.8g/kg和33.6g/kg。配制成10ml/kg的容积灌胃给药,一日2次,连续给药4周。在末次给药后2h,检测各组大鼠的血流动力学指标:HR、LVSP、LVEDP、LV dp/dt min和LV dp/dt max;并检测心肌组织AMPK和FFA含量。结果与假手术组比较,模型组大鼠HR、LVSP、LVEDP、LV dp/dt min升高和LV dp/dt max降低,心肌组织FFA含量增高,AMPK含量降低,差异有统计学意义(P<0.05);与模型组比较,SSQXL、SSQXM和SSQXH组大鼠HR、LVSP、LVEDP、LV dp/dt min降低,LV dp/dt max升高,FFA含量降低,AMPK含量增高,差异有统计学意义(P<0.05);与SSQXM组比较,SSQXL组大鼠HR、LVSP、LVEDP、LV dp/dt min升高和LV dp/dt max降低,FFA含量增高,AMPK含量降低,差异有统计学意义(P<0.05);SSQXH组大鼠HR、LVSP、LVEDP、LV dp/dt min降低,FFA含量降低,AMPK含量增高,差异有统计学意义(P<0.05)。结论双参强心方可改善心肌梗死后心衰模型大鼠心脏功能,提高心肌组织AMPK含量,降低FFA含量,有效调节心衰大鼠心肌能量代谢紊乱。
Objective To study the effects of Shuang Shen Qiang Xin Recipe on the hemodynamic and myocardial energy metabolism in rat with heart failure. Methods SD rats were divided into 5 groups, adopted to left anterior descending coronary artery ligation is establish the model of heart failure after myocardial infarction, control group and model group were given 0. 9% normal saline,Shuang Shen Qiang Xin low dose, middle dose and high dose group were given Shuang Shen Qiang Xin of 8.4 g/kg, 16. 8 g/kg and 33.6 g/kg. Mixture of 10 ml/kg volume to fill the stomach, 2 times a day, for 4 weeks. After 2 hours in the last treatment, detection hemodynamic parameters each groups of rats: HR, LVSP, LVEDP, LV dp/dt min and LV dp/dt Max;And detection of myocardial tissueAMPK and FFA content. Results Compared with control group, model group rats HR, LVSP, LVEDP, LV dp/dt min increased and LV dp/dt Max reduced, myocardial tissue FFA content increased, AMPK levels decreased, the difference was statistically significant (P 〈 0. 05) ; Compared with model group, SSQXL, SSQXM and SSQXH group rats HR, LVSP, LVEDP, LV dp/dt min reduced, LV dp/dt Max increased, FFA levels reduced, AMPK levels increased, the difference was statistically significant (P 〈 0. 05 ) ; Compared with SSQXM group, SSQXL group rats HR, LVSP LVEDP, LV dp/dt min increased and LV dp/dt Max reduced, FFA content increased, the AMPK content reduced, the difference was statistically significant (P 〈0.05) ; SSQXH group rats HR, LVSP, LVEDP, LV dp/dt rain reduced, FFA content reduced, AMPK content increased, the difference was statistically significant (P 〈 0.05). Conclusion Shuang Shen Qiang Xin Recipe can improved cardiac function with heart failure after myocardial infarction rats model, increased the myocardial tissue content of AMPK, reduce the content of FFA, regulate myocardial energy metabolic disorders in heart failure rat.
