摘要
为了研究陆川猪HSP27基因的遗传变异情况,试验采用RT-PCR和克隆方法,扩增了HSP27基因的编码区序列,并进行了测序和序列分析及其蛋白质结构预测。结果表明:陆川猪HSP27基因编码区全长624 bp,编码207个氨基酸;与Gen Bank中已发表的猪HSP27基因序列相比,核苷酸有1个位点发生碱基突变,但并未引起相应氨基酸的改变;陆川猪HSP27成熟肽包含α螺旋、β转角、延长线和无规卷曲;陆川猪HSP27基因与野猪的亲缘关系最近。
To study the genetic variation of the heat shock protein 27 ( HSP27 ) gene in Luchuan pig, the coding region sequence of HSP27 gene in Luchuan pig was amplified using RT - PCR and a cloning method, and then the sequencing, sequence analysis and the prediction of protein structure of HSP27 were carried out. The results showed that the full - length of the coding region of the HSF27 gene in Luchuan pig was 624 bp, encoding for a protein of 207 amino acids. Compared with the HSP27 gene sequences pubiished in GenBank, a base mutation was detected at one nucleotide site, but it did not cause the change in the corresponding amino acid. The HSP27 mature peptide in Luchuan pig contained α helix, β comer,extension cords and random coils. The Luchuan pig had the closest relationship with Sus scrofa in the HSP27 gene.
