摘要
根据齿兰环斑病毒外壳蛋白基因序列,设计了LAMP检测引物,并结合纳米磁珠技术对齿兰环斑病毒进行LAMP检测。结果表明,只有齿兰环斑病毒毒株有扩增,而建兰花叶病毒、烟草花叶病毒和黄瓜花叶病毒毒株都没有扩增,说明本实验设计的LAMP检测引物具有检测齿兰环斑病毒的特异性。灵敏度实验结果表明,纳米磁珠-LAMP结合技术比RT-PCR方法高100倍。研究表明,纳米磁珠-LAMP结合技术检测方法是一种特异、灵敏、快速的齿兰环斑病毒检测方法。
Specific LAMP primers were designed basing on the Odontoglossum ringspot virus CP gene sequence in Gen Bank.The primers used in this assay were shown to be specific for ORSV and did not react with Cym V,TMV and CMV strains.The detection limit of the method was higher about 100 times than that of the RT-PCR method.So the method of LAMP combining with MNP technology is a specific,sensitive,rapid and highly effective method for the detection and identification of ORSV.
