黄连素通过上调Smad7减轻2型糖尿病大鼠肾脏损伤 被引量:13

Renoprotective Effect of Berberineon Kidney Injury of Type 2 Diabetic Rat Modelvia Upregulating Smad7

摘要 目的:观察黄连素对2型糖尿病大鼠肾脏损伤的保护作用,并探讨可能机制。方法:将大鼠分为正常对照组、糖尿病模型组以及黄连素治疗组,给药20周后检测血糖、24 h尿蛋白,观察肾脏病理改变,并检测炎症(IκBα)及纤维化(TGF-β1、TβRI、Smad7)信号通路。结果:与正常对照组比较,糖尿病大鼠肾重指数增加,24 h蛋白尿维持在高水平,肾脏病理显示肾小球系膜基质增生,肾小管扩张变性且排列紊乱,小管间质存在炎症浸润;黄连素治疗显著降低模型大鼠肾重指数及24 h蛋白尿,减少肾皮质TGF-β1、TβRI表达,上调Smad7和IκBα表达。结论:黄连素对2型糖尿病大鼠具有肾脏保护作用,机制可能是通过上调Smad7表达从而改善肾脏炎症及纤维化。 Objective: To observe the renoprotective effect and mechanisms of berberine on a type 2 diabetes rat model.Methods: Wistar rats were randomly divided into normal control group,diabetic group and berberine- treated group. Rats were sacrificed 20 weeks later. We detected the kidney weight index,24 h urine protein,morphology study,inflammationand fibrosis markers,such as IκBα,TGF- β1,TβRI and Smad7. Results: Compared with normal rats,the diabetic rats showed higher kidney weight indexand 24 h urine protein,severe morphological changes; berberine treatment decreased kidney weight index and 24 h urine protein,alleviatedmorphological changes,suppressed the expression of TGF- β1,TβRI,meanwhile increased the expression of IκBα and Smad7. Conclusion: Berberine has renoprotectiveeffect on a type 2 diabetic rat model. The mechanism might suppressinflammation and fibrosis via upregulating expression of Smad7.
