摘要
为确定四川某猪场导致猪细菌性感染死亡的病原,本研究从送检病死猪肺脏样品中分离得到一株细菌,对该菌株进行培养特性观察、生化试验、动物致病性试验、药敏试验、16S r RNA基因的扩增及测序鉴定。结果显示其16S r RNA基因序列与肺炎克雷伯菌(Kpn)序列同源性达99%,表明该菌株为Kpn肺炎亚种;致病性试验结果显示其对小鼠具有强致病性;并且在血平板上出现α溶血现象;药敏试验结果显示分离菌株表现出多重耐药性,毒力较强,并且在培养特性上发生一定的改变。本实验为进一步研究Kpn耐药机制和致病机理奠定了基础。
To determine the pathogenic bacteriain the effected pigs, a bacteria was isolated from the lung of diseased pig and identified as gAebsiella pneumoniaeby biochemical tests, pathogenicity test, and bacteria 16S rRNA gene sequencing, which was 99% homology with K.pneumoniae. In addition, bacterial colonies were formed on blood agar, presenting sight a hemolytic phenomenon. Results of drug sensitivity tests show that the bacterial isolate was only sensitive to gentamicin, amikacin and ceftizoxime; While, resistant to cefoperazone, piperacillin, amoxicillin+clavulanicacid, tetracycline and sulfamethoxazole etc in the 19 tested drugs. In conclusion, the K.pneumoniae isolated from swine lungs had not only altered in cultural properties, but also has a strong virulence and a serious multi-drug resistance. The isolate would faciliate further the resistance mechanisms and pathogenesis-related research.
