摘要
为查明中山市某鸽场病鸽的病因,进而制定合理的治疗方案,对中山市某鸽场的肉鸽进行了病理剖检,细菌分离、培养、纯化,特性观察,镜检,16S rDNA测序,序列分析及构建进化树判断疑似致病菌。方法:利用家禽生理解剖技术对肉鸽进行解剖,培养肝脏及粪便中所分离的疑似致病菌。对疑似病原菌进行革兰氏染色、基因组DNA提取、PCR扩增及测序,进而进行同源性比对,选择合适的菌株构建系统发育树。结果:从病鸽肝脏分离到一株疑似病原菌20210801.OX1,革兰氏染色呈红色,DNA跑电泳的结果显示在1500bp,根据测序结果构建进化树的结果初步判断肉鸽分离的疑似致病菌为弗氏志贺氏菌。
In order to find out the etiology of diseased pigeons in a large-scale pigeon farms in Zhongshan and formulate reasonable treatment plans,the pigeons in pigeon farms in Zhongshan were examined by pathology,bacteria isolation,culture,purification,characteristics observation,microscopy,16S rDNA sequencing,sequence analysis and the construction of evolutionary tree to identify suspected pathogenic bacteria.Method:The suspected pathogenic bacteria isolated from liver and feces of pigeon were cultured by anatomical technique of poultry physiology.Gram-staining,genomic DNA extraction,PCR amplification and sequencing were performed on the suspected pathogens,and then homology was performed to select appropriate strains to construct phylogenetic trees.Results:a suspected pathogen 20210801.OX1 was isolated from the liver of the diseased pigeon.The gram staining was red,and the DNA electrophoresis results showed that the strain was 1500 bp.According to the sequencing results of constructing the evolutionary tree,the suspected pathogen isolated from the pigeon was shigella fosteri.
