摘要
目的探讨虫类中药全蝎、蜈蚣水煎剂对小鼠Lewis肺癌的抑制作用及其对免疫器官的影响。方法复制C57BL小鼠Lewis肺癌模型并随机分为8组(每组10只):全蝎高、中、低剂量组(1600,800,400 mg·kg-1),蜈蚣高、中、低剂量组(1200,600,300 mg·kg-1),环磷酰胺组(20 mg·kg-1)和氯化钠对照组,分别每日给予0.2m L相应药液腹腔内注射,共14 d。其中,全蝎和蜈蚣中剂量组的剂量约相当于成人每日4 g和3 g的常规用量。实验结束时处死动物,记录体质量、瘤质量、脾脏及胸腺质量,分析、比较各组的瘤质量及脾脏指数、胸腺指数。结果全蝎、蜈蚣各剂量组均有一定的抑瘤作用,抑瘤效应与剂量相关,随着药物剂量的增大,抑瘤率相应增高。其中,全蝎高、中剂量组和蜈蚣高剂量组的抑瘤作用较为明显(抑瘤率分别为36.5%、29.0%和27.5%),3组瘤质量均明显低于氯化钠对照组(P<0.05,P<0.01);各剂量组平均瘤质量(除全蝎高剂量组外)均明显高于环磷酰胺组(P<0.001);全蝎低剂量组和蜈蚣中、低剂量组虽有一定的抑瘤作用(抑瘤率分别为20.0%、18.0%、14.0%),但平均瘤质量与氯化钠对照组比较无统计学意义(P>0.05);在脾脏指数和胸腺指数方面,环磷酰胺组及全蝎高、中剂量组均明显低于氯化钠对照组(P<0.05,P<0.01)。结论全蝎、蜈蚣水煎剂对小鼠Lewis肺癌的生长有一定的抑制作用,且抑瘤效应与剂量之间有一定的相关性,其中以全蝎高、中剂量组和蜈蚣高剂量组的抑瘤作用较为明显,全蝎高剂量组最为明显。全蝎高、中剂量组及环磷酰胺组对小鼠脾脏和胸腺有较为明显的抑制效应,而全蝎低剂量组及蜈蚣各剂量组则无明显影响。
Objective To investigate the inhibitory effect of Chinese medicine scorpion and centipede decoction on Lewis lung cancer in mice, and to observe its effect on the immune organs. Methods C57 BL mice model of Lewis lung cancer was established and then the mice were evenly randomized into 8 groups, namely cyclophosphamide group(20 mg·kg^-1),sodium chloride control group,high-,moderate- and low-dose scorpion groups(in the dosage of1600, 800, 400 mg·kg^-1, respectively), and high-, moderate- and low-dose centipede groups(1200,600,300 mg·kg^-1,respectively),10 rats in each group. The mice were orally given the corresponding medicine in0.2 m L for 10 days according to the experimental design. The middle dose of scorpion and centipede was the equivalent dose of 4 g,3 g per day for the adult respectively. At the end of experiment,the mice were executed. The body mass,tumor mass,spleen index and thymus index were recorded and compared. Results Scorpion and centipede in different dosages showed certain antitumor effect,and the effect was dose-dependent. High- and middle-dose scorpion groups and high-dose centipede group had obvious antitumor effect,and the tumor inhibition rate was 36.5 %,29.0 % and27.5 %,respectively,higher than that of the sodium chloride control group(P0.05). Except the high-dose scorpion group(P〈0.05), the other medication groups had higher average tumor mass than cyclophosphamide group(P〈0.001). The tumor-inhibition rate of low-dose scorpion group,and middle- and low-dose centipede groups was20.0 %,18.0 %,14.0 %,respectively, but their average tumor mass did not differ from that of sodium chloride control group(P〈0.05). Cyclophosphamide group and high- and middle-dose scorpion groups had lower spleen index and thymus index than sodium chloride control group(P〈0.05). Conclusion Scorpion and centipede decoction could dose-dependently inhibit the growth of Lewis lung cancer in mice,and high- and middle-dose scorpion groups and high-dose centipede group have obvious antitumor effect. Scorpion in high and middle dosage and cyclophosphamide show more obvious inhibitory effect on the mice spleen and thymus,but low dose of scorpion and centipede have no significant influence.
