摘要
目的检测慢性乙型肝炎患者外周血中Treg细胞的比例,并探讨其对DC细胞的免疫抑制作用。方法抽取慢性乙型肝炎(CHB)患者和正常对照组的外周血,流式细胞术检测CD4+CD25+Treg细胞的比例以及DC细胞表面CD80和人白细胞(位点)DR抗原(HLA-DR)的表达;在DC培养的不同时间内加入不同比例CHB患者的Treg细胞,检测DC对淋巴细胞增殖功能的影响;检测DC培养的不同时间内加入不同比例CHB患者的Treg细胞后培养上清中细胞因子白细胞介素-10(IL-10)和肿瘤转化因子-β(TGF-β)的表达。结果正常人外周血中Treg(CD4+CD25+Foxp3+)细胞的比例为(4.12±1.36)%,CHB患者外周血中Treg细胞的比例为(10.54±3.27)%,CHB患者外周血中的Treg细胞比例明显高于正常组,差异有统计学意义;CHB患者表面分子CD80的表达为(60.12±5.23)%,HLA-DR的表达为(61.21±4.28)%;而正常组的CD80的表达为(84.13±4.33)%,HLA-DR的表达为(86.57±3.82)%,CHB患者DC细胞表面分子CD80和HLA-DR的表达均显著低于正常组,差异有统计学意义;经Treg细胞处理后,DC细胞刺激淋巴细胞的增殖能力下降,淋巴细胞增殖的抑制率较正常组均显著升高;Treg细胞与DC细胞比例为1∶1、1∶10以及1∶100时,IL-10和TGF-β的表达均显著高于正常组,并且随着Treg细胞的比例增高,细胞因子的水平也相应增高,差异有统计学意义。结论 CHB患者外周血中Treg细胞比例增高,可能是通过诱导IL-10和TGF-β的表达抑制DC细胞的免疫功能。
【Objective】To investigate the immunosuppressive effect of the regulatory T(Treg) cells on the dendritic cells in the pathogenesis of chronic hepatitis B(CHB).【Methods】Peripheral blood mononuclear cells(PBMC)were isolated from CHB patients and normal people, and flow cytometry were used to detect the frequency of CD4^+CD25^+Treg cells and the expression of CD80 and HLA-DR on the surface of dendritic cells(DCs); Treg cells from CHB group and DCs from normal group were cocultured under different ratio and time, and the suppressing index was detected; coculture supernatant concentrations of interleukin-10(IL-10) and tumor transforming factor-β(TGF-β) were detected by enzyme linked immunosorbent assay(ELISA). 【Results】The frequency of Treg cells in CHB group was(10.54 ± 3.27)%, and its frequency in normal group was(4.12 ± 1.36)%, and the difference had statistical significance; the expression of CD80 on the surface of DCs from CHB group was(60.12 ± 5.23)%, while the expression of HLA-DR on the surface of DCs from CHB group was(61.21 ± 4.28)%; the expression of CD80 in normal group was(84.13 ± 4.33)%, while the expression of HLA-DR in normal group was(86.57 ± 3.82)%; the expression of CD80 and HLA-DR on the surface of DCs in CHB patients was significant lower than that in normal group; CD4^+CD25^+Treg cells from CHB group had significant suppressing effects on the immune function of DCs which showed time and dose dependence; the secretion of IL-10 and TGF-β increased significantly in CHB group.【Conclusions】The increasing proportion of Treg cells in peripheral blood of patients with CHB may suppressing the immune function of DCs by inducing IL-10 and TGF-β expression.
