急性髓系白血病患者循环microRNA-335的检测及其临床意义 被引量:4

摘要 目的探讨急性髓系白血病(AML)患者循环miR-335的表达及其临床意义。方法本研究共纳入26例初诊AML患者和26例健康对照者,取外周血通过实时荧光定量PCR检测miR-335表达量并分析其与AML临床资料的相关性。绘制ROC曲线、计算曲线下面积以评估miR-335对新发AML的诊断价值。结果循环miRNA-335相对表达量在同组内纳入研究对象的年龄、性别比较上未见明显差异(P〉0.05)。与AML患者外周血白细胞计数及原始细胞计数无显著相关性(P〉0.05)。与骨髓原始细胞计数及FAB分型无显著相关性(P〉0.05)。当以相对表达量2.7为cutoff值时,miRNA-335诊断AML的AUC可达0.915(95%可信区间:0.828~1.000;P〈0.0001),miRNA-335诊断AML的敏感性可达80.8%,特异性可达100%。结论血清循环miR-335表达量在AML患者中显著升高,可作为AML辅助诊断标志物。 Objective To investigate the expression and clinical significance of circulating miR-335 in acute myeloid leukemia (AML) patients. Methods Twenty-six newly diagnosed AML patients and 26 healthy controls were enrolled in our study.The expression of miR-335 in periphral blood of AML patients was detected by Real-time quantitative PCR and its correlation with clinical material of AML was analyzed. Then the receiver operating characteristic curve (ROC) was conducted, and the area under the curve (AUC) was used to evaluate the diagnostic accuracy of miR-335 as a potential biomarker in the new AML. Results There was no significant difference of the relative expression level of circulating miR-335 with the subjects age and sex within the same group (P〉0.05). There was no significant difference of the white blood cell count and primitive cell count in periphral blood of AML (P〉0.05), the same as the primitive bone marrow cell count and FAB classification in AML (P〉0.05). ROC curve analyses revealed an AUC of 0.915 in diagnosing AML. When relative expression 2.7 used as the cut off value of miR-335, the sensitivity and specificity of miR-335 in diagnosing AML could reached 80.8% and 100%. Conclusion The expression of circulating miR- 335 significantly increased in AML patients, which could be an auxiliary diagnostic biomarker for AML.
