焦虑症海马神经元损伤分子生物学机制及丹栀逍遥散干预的研究进展 被引量:12

Advances in Mechanisms of Molecular Biology of Danzhi Xiaoyao San in Anti Hippocampal Neuronal Damage of Anxiety Disorder

摘要 关于焦虑症准确的发病机制目前研究比较一致地认为存在大脑海马神经元的损伤,国外研究指出该损伤与蛋白磷酸酶2A(protein phosphatase 2A,PP2A)的调控、α-突触核蛋白(α-synuclein,α-syn)的表达有重要关系,因此笔者认为PP2A活性的改变,可继发α-syn异常表达,引起神经元的相关改变;那么通过调控PP2A活性,可能引起α-syn蛋白C端129位的丝氨酸磷酸化/去磷酸化,从而调控GR活性,抗海马神经元凋亡,达到修复受损海马,抗焦虑的作用。本文综述了近年有关蛋白磷酸酶2A,α-突触核蛋白在焦虑症的分子生物学机制的研究进展和中医药复方丹栀逍遥散治疗焦虑症机制的实验研究进展。为临床有效治疗焦虑症开辟新道路。 It was widely accepted, in the present studies on the exact pathogenesis of anxiety disorders, that there is injury of hippocampal neurons in brain. Overseas studies indicate that injury play an important role in the regulation of protein phosphatase 2A (protein phosphatase 2A, PP2A) and the expression of α-synuclein (α- synuclein). Therefore, we consider that the change in the activity of PP2A may induce the abnormal expression of α-syn, causing relative changes in neurons; so the phosphorylation/dephosphorylation of 129 points α-syn protein C-terminal9 might be caused through regulating the activity of PP2A, then GR activity against neuronal apoptosis are further regulated, to repair damaged hippocampus, then to work in the anti-anxiety disorder. This paper reviews the research progress on the protein phosphatase 2A, α-synuelein in the molecular mechanisms of anxiety disorders and the progress on experimental study of the mechanism of Chinese medicine compound Danzhi Xiaoyao San in the treatment of anxiety disorders in recent years.
