摘要
以蔗髓为原料、亚硫酸氢镁法制浆废液为胶黏剂制备的蔗髓缓冲包装材料为研究对象,从土壤中筛选出其高效降解菌株,并进行鉴定,确定菌株的稳定性。采用平板水解圈法和失重法进行菌株筛选,同时采用扫描电子显微镜(SEM)对蔗髓缓冲包装材料(BPCPMs)降解前后的形貌进行表征。利用培养特征、形态学、分子生物学方法进行菌株鉴定。筛选出效果稳定的蔗髓缓冲包装材料高效降解菌株ZS0918,经基因鉴定该菌株为杂色曲霉(Aspergillus versicolor)。SEM结果表明,在霉菌作用下,BPCPMs表面出现大小不一的孔洞,材料变薄,发生显著降解。
The screening BPCPMs degrading microorganisms was studied. The methods used to screen the effective strains included hydrolysis spot diameter measurement of hemicellulose plate, weight-loss method, and scanning electron microscope. Methods used to identify the strains included culture characteristics, morphological, physiological-biochemical characteristics and molecular biological methods. Isolated 5 moulds with higher degrading ability. To the best one of degradability, made hemicellulose degrading activity testing from liquid submerged-culture, up to 37.5 U/mL. By molecular biological methods, ZS0918 was Aspergillus versicolor.
