氟尿嘧啶诱导大鼠卵巢损伤模型 被引量:1

Establishment of Rat Model of Fluorouracil-induced Premature Ovarian Damage

摘要 目的探讨氟尿嘧啶(5-Fu)诱导的大鼠卵巢损伤模型建立的方法和时间。方法将8-9周SD大鼠分为4组:生理盐水对照组、5-Fu低剂量组、中剂量组、高剂量组,各组分别接受生理盐水和5-Fu:每天1 mL/kg、5 mg/kg、10 mg/kg、20 mg/kg腹腔注射,连续14 d。通过大鼠动情周期、血清AMH、FSH和E2的水平和卵泡计数情况来判断建模效果。结果低、中、高剂量组大鼠动情周期出现紊乱。停药后第1天,与生理盐水对照组相比,血清AMH浓度低、中、高剂量组明显降低(P〈0.05);中、高剂量组血清雌二醇降低(P〈0.05);低、中、高剂量组血清FSH浓度明显升高(P〈0.05);低、中、高剂量组卵巢湿重减少明显(P〈0.05),生长卵泡和原始卵泡计数明显减少(P〈0.05)。结论 5-Fu能够损伤大鼠的卵巢功能,引起血清E2、FSH和AMH水平的改变,且在反应卵巢损伤时AMH可能较FSH和E2更灵敏。 Objective To investigate the applicable method and optimal time in establishment of the animal model for fluorouracil-induced ovarian damage. Methods 8-9week-old SD female rats were divided into four groups: normal saline control,low-dose 5-Fu,middle-dose 5-Fu,high-dose 5-FU. The rats in four groups were received daily intraperitoneal injection of normal saline and 5-Fu,1 mL / kg,5 mg / kg,10 mg / kg,20 mg / kg,respectively for two weeks. We detected vaginal smears,wet ovaries wet weighted,and the levels of estradiol( E2),follicle-stimulating hormone( FSH) and anti-Müllerian hormone( AMH) in blood to value the effect of 5-Fu. Results The estrus cycle of rats was remarkably disordered in the BCD groups. It suggested that,compared to A,the level of AMH was significantly decreased(( P〈0. 05) in groups BCD,the level of E2 was also decreased( P〈0. 05) in groups CD. While the concentration of FSH was heavily increased after 5-FU administration( P〈0. 05). The wet weight and follicle numbers were also decreased( P〈0. 05). Conclusion 5-Fu can damage the ovarian and changes the levels of AMH,E2,and FSH. AMH may be more sensitive than E2 and FSH in response to ovarian damage.
