摘要
分别采用溶剂浸提法、索氏回流法、超声提取法及微波提取法提取葡萄梗中的多酚物质,并利用高效液相色谱技术分析葡萄梗中多酚物质的组成及其含量。测定的加标回收率在94.5%~98.5%,RSD在1.3%~2.1%,该方法的准确度及精密度较好。结果表明,葡萄梗中含量较高的多酚成分为绿原酸(5.70 mg/g^13.24 mg/g)和白藜芦醇(2.22 mg/g^6.71 mg/g),微波提取法提取5 min的葡萄梗中多酚物质的效果较好,本研究可为葡萄梗多酚物质的提取提供参考。
Polyphenols of Grape stems were extracted with the methods of organic solvent extraction, soxhlet extraction, ultrasonic extraction and microwave extraction. After optimizing mobile phase, contents of polyphenols were detected by High Performance Liquid Chromatography (HPLC), equation of linear regression of seven standard material of Polyphenols were determined. The recovery rate was from 94.5 % to 98.5 % and the relative standard deviation (RSD) was from 1.3 % to 2.1% ,the method was accurate and sentitive. The contents of chlorogenic acid (5.70 mg/g-13.24 mg/g)and resveratrol (2.22 mg/g-6.71 mg/g)were higher of all the components. The method of microwave extraction for 5 min exhibited better extraction efficiency which would be used for extraction of polyphenols.
