摘要
目的探讨野艾蒿提取物对肝癌细胞系Hep G2增殖的影响及对肝癌细胞系Hep G2细胞内Bcl-2、Bax及Caspase-3基因表达的影响。方法给予不同浓度的野艾蒿提取物(0,200,400,800μg/m L)处理肝癌细胞系Hep G2 24,48,72 h,采用MTT法检测不同浓度的野艾蒿提取物对肝癌Hep G2细胞增殖的影响;用RT-PCR法检测肝癌细胞Hep G2中Bax、Bcl-2以及Caspases-3基因的表达情况。结果给予野艾蒿提取物400,800μg/m L处理72h后,Bax表达水平明显低于0μg/m L浓度(P<0.05);给予野艾蒿提取物800μg/m L处理24,48,72 h后,Bcl-2、Caspases-3基因表达水平均明显低于0μg/m L浓度(P均<0.05)。结论野艾蒿提取物对肝癌细胞系Hep G2的增殖有一定的抑制作用,其机制是野艾蒿提取物可抑制Bcl-2基因表达,上调Bax、Caspases-3基因的表达。
Objective It is to approach the influence of extractive from Artemisia lavandulaefolia DC on hepatocelular car- cinoma HepG2 cells proliferation and the expression of Bax, Bcl - 2, Caspases - 3. Methods The HepG2 cells were treated with different concentrations (0, 200, 400, 800 μg/mL) of the extractive from Artemisia lavandulaefolia DC for 24, 48 and 78 hours; then the influence on cell proliferation of different concentrations of Artemisia lavandulaefolia DC were detected by MTT; the expression of Bax, Bcl - 2, Caspases - 3 gene in HepG2 cells were detected by RT - PCR. Results After treated with 400, 800μg/mL of extractive form Artemisia lavandulaefolia DC for 72 hours, the expression of Bax gene was higher than 0μg/mL group significantly (P 〈 0.05 ). After treated with 800 μg/mL of Artemisia lavandulaefolia DC for 24, 48 and 72 hours, the expression of Bcl - 2 and Caspases - 3 gene were lower than 0μg/mL group ( all P 〈 0.05 ). Conclusion The ex- tractive of Artemisia lavandulaefolia DC have certain inhibitory effect on the proliferation of hepatoma HepG2 cells, the mecha- nism is to inhibit the expression of Bcl - 2 and up-regulation the expression of Bax, Caspasese - 3.
