摘要
选择5株蛹虫草菌株感染家蚕5龄第3天幼虫生产蚕蛹虫草,通过观察检测蚕蛹虫草的形态性状及采用反相高效液相色谱(RP-HPLC)法测定虫草素和腺苷的含量,为筛选高产优质蛹虫草菌株提供依据。不同菌株栽培生产的蚕蛹虫草的外观形状、生长状况以及活性成分含量均有明显差异:1-Y菌株栽培的蚕蛹虫草整体形状较接近于野生的冬虫夏草,MS菌株栽培的蚕蛹虫草子座形状较佳,17-3菌株栽培的蚕蛹虫草僵虫体形状较佳;1-1菌株栽培的蚕蛹虫草总质量和僵虫体质量均最高,干质量分别达到1.460 g和1.170 g;MS菌株栽培的蚕蛹虫草子座总数量、子座总长度及子座质量占虫草总质量的比率等指标均极显著高于其他菌株(P<0.01),分别达15.100根、44.877 cm和41.357%;1-Y菌株栽培的蚕蛹虫草整体所含虫草素最高,达8.92 mg/g,MS菌株栽培的蚕蛹虫草整体所含腺苷最高,达2.22 mg/g,并且5株供试菌株栽培的蚕蛹虫草僵虫体中的虫草素含量均高于子座,而子座中的腺苷含量均高于僵虫体。研究结果表明,蛹虫草菌株是影响人工栽培蚕蛹虫草形态性状及活性成分含量的重要因素,初步认为1-Y菌株和MS菌株具有较高的生产应用价值。
In this study,we cultured aweto by inoculating 5 selected Cordyceps militaris strains on the 3rd day silkworm( Bombyx mori) larvae of the 5th instar,and observed the morphological characters of aweto and detected the contents of cordycepin and adenosine by reversed-phase high performance liquid chromatography( RP-HPLC) method to provide evidence for screening high yield and quality C. militaris strains. The results showed that there were significant differences on the appearance,growth status and active component of aweto cultured with different strains of C. militaris. The overall appearance of aweto cultured with strain 1-Y was similar to that of the wild aweto. The stroma shape of aweto cultured with strain MS and the stiff larvae of aweto cultured with strain 17-3 was better than that of the other test groups.The total weight and stiff larvae body weight of aweto cultured with strain 1-1 were the highest,having a dry weight of 1. 460 g and 1. 170 g respectively. The total stroma number,total stroma length,and stroma weight percentage of aweto cultured with strain MS were extremely significant higher than those cultured with the other strains( P0. 01),being 15. 100,44. 877 cm and41. 357% respectively. The content of cordycepin in aweto cultured with strain 1-Y was the highest,reaching8. 92 mg / g. The content of adenosine in aweto culturedwith strain MS was the highest,reaching 2. 22 mg / g. The content of cordycepin in stiff larvae infected with 5 tested strains was all higher than that of stroma,whereas the content of adenosine in stroma was higher than that in stiff larvae.These results indicate that Cordyceps militaris strains are an important factor influencing the morphological characters and active component contents in artificially cultivated aweto. It is preliminary considered that Cordyceps militaris strains 1-Y and MS have high application value.
