蝙蝠葛碱对阿尔茨海默病转基因细胞模型氧化应激损伤保护作用的研究 被引量:1

摘要 目的探讨蝙蝠葛碱对阿尔茨海默病(AD)转基因细胞模型氧化应激损伤的保护作用。方法将质粒APPswe转染入SH-SY5Y细胞中,构建过表达APP的AD细胞模型,检测不同蝙蝠葛碱浓度下细胞存活率、丙二醛(MDA)含量及COX-2蛋白表达的差异。结果随着给药浓度的增加,细胞存活率逐步增加,MDA含量逐步降低,其中空白对照组、20或10倍稀释蝙蝠葛碱预处理组与模型组差异有统计学意义(P<0.05或P<0.01),10倍稀释蝙蝠葛碱预处理组与100、50倍稀释蝙蝠葛碱预处理组差异有统计学意义(P<0.05或P<0.01);COX-2蛋白表达量随着给药浓度增加而逐渐下降。结论蝙蝠葛碱对AD转基因细胞模型的氧化应激损伤具有一定的抑制作用,且呈现出剂量效应。 Objective To explore the protective function of dauricine on oxidative stress inju- ry induced by APP over-expressing in Alzheimer's disease (AD) cell model. Methods APPswe plas- mid was transfeeted into SH-SYSY cells in order to build APP over-expressing AD cell model. Differ- ences of cell survival rate, malondialdehyde (MDA) and COX-2 protein expression with different con- centrations of daurieine were analyzed. Results The survival rate of cell increased and MDA content decreased gradually with the increase of medication concentration, and there were significant differ- ences between the model group and control group, 20 or 10-fold dilution daurieine pretreated group (P 〈0.05 or P 〈0.01 ). There were significant differences between 10-fold dilution daurieine pretreat- ment group and 100-fold or 50-fold dilution daurieine pretreatment group (P 〈 0.05 or P 〈 0.01 ). COX-2 protein expression decreased gradually with increasing of medication concentration. Conclusion Dauricine shows a dose effect of protection on oxidative stress induced by APP over-expressing in AD gene transfected cell model.
