摘要
目的:观察大鼠黑质内注射脂多糖(LPS)后黑质环氧化酶-2(COX-2)蛋白表达和前列腺素E2(PGE2)水平的变化。方法:大鼠黑质立体定向注射15μgLPS后不同时间点,免疫组化方法观察黑质酪氨酸羟化酶(TH)阳性神经元数量的变化,免疫印迹法检测黑质COX-2蛋白表达以及放射免疫方法测定PGE2水平。结果:LPS注射后TH阳性细胞数量24h显著减少,14d达到高峰;黑质COX-2蛋白表达6h开始增高,14d达高峰,在30d仍高于对照组。而PGE2水平虽然增加,但并不和COX-2蛋白表达平行。结论:COX-2可能涉及LPS诱导的黑质多巴胺神经元的变性过程。
Objective:To investigated the changes of cyclooxygenases-2(COX-2) protein expression and PGE2 content in the substantia nigra(SN) following intranigral injection of lipopolysacchride(LPS).Methods:At the different survial time(6h,24h,14d and 30d) after injection of LPS in the SN of rats,the number of tyrosine hydroxylase(TH)-positive neurons were observed by immunohistochemistry,COX-2 protein expression by immunoblotting,PGE2 production was measured with enzyme linked immunoassay.Results:The number of TH-positive neurons is not changed on the 6 hour,but the significant loss were observed on the 24h and the maximum of loss on the 14d;COX-2 protein significantly increased from 6h after LPS injection achieving maximum on the 14d,however,it persisted and elevated until the 30d.PGE2 productions in the SN and serum of rat increased also,but did not followed the time pattern of COX-2 protein expression.Conclusion:COX-2 may be involved in dopaminergic neurons degeneration induced by LPS.
