摘要
目的观察糖肝康对糖尿病脂肪肝大鼠肝组织cAMP反应元件结合蛋白(CREB)表达的影响,探讨其作用机制。方法采用链脲佐菌素联合高脂饮食复制糖尿病脂肪肝大鼠模型。造模成功后随机分为模型组、护肝宁组和糖肝康小、中、大剂量组,每组10只,另取10只正常大鼠作为空白组。各给药组给予相应药物干预,12周后采用Western blot检测大鼠肝组织CREB的表达。结果与空白组比较,模型组肝组织CREB表达明显增高(P<0.01);与模型组比较,各给药组肝组织CREB表达明显减少(P<0.01),糖肝康中、大剂量组优于护肝宁组(P<0.01)。结论糖肝康可能通过降低CREB表达水平改善肝脏代谢,进而保护肝脏。
Objective To observe the effect of Tanggankang on the expression of CREB indiabetic-fatty liver rats;To investigate the mechanism of Tanggankang. Methods STZ and feedinghigh fat forage induced diabetic rats were randomly divided into model group, Huganning group,Tanggankang high dose group, Tanggankang medium dose group, Tanggankang low dose group,10 rats in each group, with 10 normal rats as control. After 12 weeks of drug intervention, ratswere killed, the livers were removed, and the expression of CREB was detected by Western blot.Results Compared with normal group, the level of CREB of model group was significantly decreased（P 〈 0.01）. Compared with model group, Tanggankang low, medium and high dose groupsignificantly decreased the level of CREB （P 〈0.01）. Conclusion Tanggankang may play an activerole in glucose and lipid metabolism by affecting the expression of CREB.
