产多种活性物质的噬几丁质属菌株ZLX3的筛选及鉴定 被引量:3

Screening and identification of Chitinophaga ZLX3 that can produce various active substances

摘要 目的筛选可产生多种生物活性物质的噬几丁质属Chitinophaga菌株。方法采用VY/4平板法从兔粪、羊粪、泥土、朽木等样品中筛选目的菌株;用滤纸片法检测抗菌活性;用纤维蛋白平板法检测溶栓活性;用纤维蛋白原平板法检测抗凝活性;用形态学特征、理化性质、16S rRNA序列同源性鉴定目的菌株。结果从朽木样品中分离到一株可同时产生广谱抗生素和抗血栓活性物质的ZLX3菌株,其分泌的胞外液具明显的广谱抗菌作用和溶栓抗凝活性;结合形态学、理化性质、16S rRNA序列同源性及进化树分析,发现ZLX3菌株属于Chitinophaga属,将其命名为Chitinophaga sp.ZLX3。结论 ZLX3菌株可产生广谱抗生素和抗凝溶栓活性物质。 OBJECTIVE To screen the Chitinophaga strains that can produce various bioactive substances. METHODS The target strains was screened on VY/4 plate from dozens of samples such as rabbit manure, sheep manure,soil,deadwood, etc. The antibiotic activity of the target strains was detected by filter paper disk method. The fibrinolytic activity of the target strains was detected by the the fibrin plate and the anticoagulant activity was detected by fibrinogen plate. The target strains was identified by morphological characteristics, physical and chemical properties and 16S rRNA sequence. RESULTS An strains ZLX3 that could produce the broad - spectrum antibiotic and antithrombotic from a rotten wood sample was obtaine. The extracellular fluid of the strains ZIX3 showed both obvious antibiotic activity and antithrombotic activity. According to the morphology, physico -chemical properties, 16S rRNA sequence homology and phylogenetic tree, the strains ZLX3 belonged to the genus Chitinophaga, and were identified as Chitinophaga sp. ZLX3. CONCLUSION The strains ZLX3 can produce obvious the broad -spectrum antibiotic and anticoagulation and thrombolysis active substance.
