重组质粒pEGFP-N1-Twist对SKOV3细胞增殖、侵袭及迁移能力的影响 被引量:3

摘要 目的评估重组质粒pEGFP-N1-Twist对卵巢癌SKOV3细胞增殖、侵袭及迁移能力的影响。方法将pEGFP-N1-Twist转染入SKOV3细胞系,用real-time PCR和Western blot检测Twist基因与蛋白的表达情况;通过Transwell及MTT试验检测pEGFP-N1-Twist对SKOV3细胞的作用。结果转染了pEGFP-N1-Twist的SKOV3细胞中Twist基因/蛋白的表达明显上调;转染组较空白对照组侵袭及迁移能力增强(P<0.05),细胞增殖能力明显增加(P<0.05)。结论成功构建了Twist基因的真核表达载体并能在细胞内稳定表达,Twist基因能增强SKOV3细胞的侵袭及迁移能力、细胞增殖能力。 Objective To explore the effects of pEGFP-N1-Twist on proliferation,invasion and migration of SKOV3 cells.Methods The pEGFP-N1-Twist was transfected into SKOV3 cells.The expression of Twist gene was detected in pEGFP-N1-Twist by real-time PCR and Western blot.The effects of Twist on migration and invasion of SKOV3 cells were examined by Transwell assay and MTT respectively.Results The SKOV3 cells transfected with pEGFP-N1-Twist plasmid expressed a higher level of Twist protein steadily.The capabilities of migration,invasion and proliferation of SKOV3 cells transected by pEGFP-N1-Twist increased significantly compared with blank control group (P < 0.05).Conclusion The recombinant plasmid pEGFP-N1-Twist can be constructed successfully and steadily expressed.Twist gene can enhance the abilities of migration,invasion and proliferation of SKOV3 cells.
