摘要
目的:探讨肠易激综合征动物的P物质(SP)及c-fos的表达变化,以及它们在IBS中的可能作用和临床意义。方法:将SD大鼠随机分为正常对照组、模型组、疏肝健脾组,给予大鼠反复直结肠灌注中药大黄造成炎症刺激,炎症恢复后予小量电刺激诱发高敏感性的肠道发生应激反应,制备IBS动物模型,通过免疫组织化学法测定对SP和c-fos表达变化及疏肝健脾法对以上指标的调控作用。结果:模型组与正常对照组比较,SP、c-fos阳性表达明显升高(P<0.01);疏肝健脾组与模型组比较,SP、c-fos阳性表达减少(P<0.01,P<0.05)。结论:大黄-电刺激大鼠模型可有效制备IBS动物模型;基于"脾主运化"理论的疏肝健脾法能够改善该模型的炎症损伤,对IBS有一定的防治作用。
Objective:To investigate the mechanism of soothing liver and invigorating spleen on substance P (SP) and c -fos expression changes on the rats models with IBS. Methods:SD rats were randomly divided into the blank control group, model group, soothing liver and invigorating spleen group. The rat models with IBS were made by perfusing rhubarb repeatedly to cause inflammation. After inflammation recovering, the rat was stimulated with weak electric current to induce the supersensitivity from hypersensitive intestine. And then we observed effects of soothing liver and invigorating spleen on the expression of SP and c - fos in colonic mucosal tissue during the stimulation. Results : Compared with the blank control group, the expressions of SP and c - fos in the colonic mucosal of the model group were increased significantly (P 〈 0. 01 ). After the treatment, the contents in the colonic mucosal were decreased (P 〈 0.01, P 〈 0. 05 ). Conclusion : The rats models with IBS can effectively simulate human irritable bowel syndrome, and the soothing liver and invigorating spleen can improve prevention and treat the symptoms of the rats models in some degrees.
