摘要
目的 :探讨小鼠 β2 m正、反义核酸表达载体pcDNA3- β2 mSN和pcDNA3- β2 mAN对NIH3T3细胞表达β2 m的影响 ,为降低MHCⅠ类分子表达的研究提供新方法和实验依据。方法 :本研究应用分子生物学技术 ,构建小鼠 β2 m正、反义核酸表达载体pcDNA3- β2 mSN和pcDNA3- β2 mAN ,用脂质体转染NIH3T3细胞。经过RT -PCR及Westernblot检测 ,观察正、反义核酸对细胞 β2 mmRNA和蛋白表达的影响。结果 :小鼠 β2 m正、反义核酸表达载体成功导入NIH3T3细胞中 ,且有效表达 ,转染正义核酸的细胞 β2 mmRNA和蛋白表达水平升高 ,而转染反义核酸则 β2 mmRNA和蛋白表达水平降低。结论 :小鼠 β2 m反义核酸表达载体pcDNA3- β2 mAN可以降低NIH3T3细胞 β2 m基因的表达 ,本研究结果为降低MHCⅠ类分子的表达。
AIM: The expression of the mouse β 2 microglobulin (β 2 m) in NIH3T3 cells transfected with the mouse β 2m sense and antisense RNA was detected to clarify the effect of mouse β 2m sense and antisense RNA on the expression of MHC classⅠgene. METHODS: The mouse sense and antisense RNA, pcDNA3-β 2mSN and pcDNA3-β 2mAN, were constructed and were transfected into NIH3T3 cells by lipofectamine. RT-PCR and Western blot were used to detect the expression of β 2m in those cells. RESULTS: The expression of the mouse β 2m in the cells transfected with pcDNA3-β 2mSN was increased, while it was decreased in those cells transfected with pcDNA3-β 2mAN. CONCLUSION: pcDNA3 -β 2mAN can downregulate the expression of the β 2m in NIH3T3 cells.
