基于血清代谢组学探究玄参对脾虚水湿不化大鼠的利水作用机制 被引量:6

Study on the Diuresis Mechanism of Radix Scrophulariae on Rats with Spleen Deficiency and Water Dampness Based on Serum Metabonomics

摘要 目的:本实验采用血清代谢组学的方法对玄参在脾虚水湿不化模型大鼠的利水作用机制进行研究,并通过寻找具有生物学意义的内源性潜在生物标志物,以此探寻其相关代谢通路。方法:研究采用高脂饲料饮食诱导的方式对大鼠进行造模并对血清蛋白、胃泌素、胃排空率及水负荷等利水相关药效学指标进行检测,应用UPLC-Q-TOF-MS技术对空白组、模型组与玄参组大鼠血清进行分析,探求其潜在的关键性生物标志物及对应的相关性代谢通路。结果:玄参可改善脾虚水湿不化大鼠相关的药效学指标,同时寻找出与脾虚水湿不化模型及给予玄参干预后具有共同差异的生物标志物29个,涉及13条相关的代谢通路,应用相关代谢通路分析发现,影响最为广泛的4条代谢通路可能是原发性胆汁酸生物合成、鞘脂类代谢、叶酸合成及甘油磷脂代谢。结论:故推测玄参对脾虚水湿不化状态下血清代谢产物具有调节作用,从而为进一步对玄参利水功效的临床安全应用提供理论依据。 The method of serum metabonomics was adopted in this experiment to study the diuresis mechanism of Radix scrophulariae on the rat models of spleen deficiency and water dampness,and the biologically significant endogenous potential biomarkers was searched to explore the related metabolic pathway.The model of rats was established by inducing with high fat diet,and the pharmacodynamics indicators such as serum protein,gastrin,gastric emptying rate and water load were measured.UPLC-Q-TOF-MS technique was used to analyze the serum of rats in normal group,model group and Radix scrophulariae group to explore the potential key biomarkers and their related metabolic pathways.The results showed that Radix scrophulariae can improve the relevant pharmacodynamic indicators of rats with spleen deficiency and water dampness,and there were 29 biomarkers which had common differences with the model of spleen deficiency and water dampness after intervention with Radix Scrophulariae were found,involving 13 related metabolic pathways.Based on the analysis of relative metabolic pathways,it was found that the primary bile acid biosynthesis,sphingolipid metabolism,folate biosynthesis and glycerol phospholipid metabolism were the most widely affected metabolic pathways.Therefore,it is speculated that Radix scrophulariae can regulate the serum metabolites under the condition of spleen deficiency and water dampness,thus providing a theoretical basis for the further clinical safe application of Radix scrophulariae.
