定痫汤对癫痫模型大鼠海马BDNF/Trk B通路的影响 被引量:8

Effects of Dingxian Decoction(定痫汤) on BDNF/TrkB Pathway of Hippocampus in Epilepsy Model Rats

摘要 目的探讨定痫汤抗癫痫的可能作用机制。方法将66只大鼠随机分为模型组、定痫汤组、定痫汤+K252a组各22只,采用锂-匹罗卡品建立大鼠癫痫模型,另将22只健康大鼠设为正常组。定痫汤+K252a组在大鼠造模前1周开始每日给予酪氨酸激酶受体B (Trk B)拮抗剂K252a侧脑室注入,每次5μl,每日1次,共1周;定痫汤组和定痫汤+K252a组在造模后1 h开始给予浓度为1. 55 g/ml定痫汤2ml灌胃,同时正常组、模型组均灌服等体积生理盐水,均每日1次,连续6周。各组大鼠中11只监测大鼠脑电图并记录大鼠癫痫自发性再发作行为的潜伏期、发作频率及持续时间,另11只检测大鼠海马脑源性神经营养因子(BDNF)蛋白表达。结果正常组发生癫痫自发性再发作0只,模型组大鼠9只,定痫汤组5只,定痫汤+K252组8只。与模型组比较,定痫汤组癫痫自发性再发作潜伏期明显延长,发作频率下降,持续时间明显缩短,海马BDNF蛋白表达降低(P <0. 01)。与定痫汤组比较,定痫汤+K252组癫痫自发性再发作的发作频率增加,持续时间延长,潜伏期缩短,BDNF蛋白表达降低(P <0. 01)。结论定痫汤可能通过调节BDNF/TrkB通路发挥抗癫痫作用。 Objective To explore the possible mechanism of anti-epilepsy effect of Dingxian Decoction(定痫汤). Methods A total of 66 rats were randomly divided into model group,Dingxian Decoction group and Dingxian Decoction + K252 a group,with 22 rats in each group. Lithium pilocarpine was used to establish rat epilepsy model,and 22 healthy rats were set as normal group. K252 a-antagonist tyrosine kinase receptor B(TrkB) was injected into lateral ventricles of rats in Dingxian Decoction + K252 a group one week before the establishment of model,5 μl each time and once a day for a week;Dingxian Decoction group and Dingxian Decoction + K252 a group were given 2 ml Dingxian Decoction through lavage at the concentration of 1. 55 g/ml 1 hour after the establishment of model,while normal group and the model group were given the same volume of normal saline once a day for 6 weeks. Eleven rats in each group were monitored by electroencephalogram(EEG) and the latency,frequency and duration of spontaneous epileptic recurrence were recorded. The expression of brain-derived neurotrophic factor(BDNF) in hippocampus was detected in the other 11 rats of each group. Results There was no spontaneous epilepsy recurrences in the normal group,while 9 rats in the model group,5 rats in the Dingxian Decoction group and 8 rats in the Dingxian Decoction +K252 group showed spontaneous epilepsy recurrence. Compared with the model group,the latency of spontaneous recurrence of epilepsy in Dingxian Decoction group was significantly prolonged,the frequency and duration of epilepsy were significantly reduced,and the expression of BDNF protein in hippocampus was decreased(P < 0. 01). Compared with Dingxian Decoction group,the frequency and duration of spontaneous epilepsy recurrence in Dingxian Decoction + K252 group were increased,and the latency was shortened and the expression of BDNF was decreased(P < 0. 01). Conclusion Dingxian Decoction may play an anti-epileptic role by regulating BDNF/Trk B pathway.
