基于“有故无陨”探讨何首乌不同成分毒性机制研究 被引量:2

摘要 目的:研究何首乌不同成分致小鼠肝损伤程度的影响。方法:将何首乌4种不同成分大黄素、大黄酚、大黄酸、鞣质按照18.9mg/kg、0.088mg/kg、0.156mg/kg、2290mg/kg剂量用羧甲基纤维素钠(CMCC)配成溶液,试验将小鼠随机分为给药组(4组),每天灌胃1次(1mL/50g),空白对照组灌胃等量蒸馏水,连续4周。实验结束,采集血清与肝脏组织标本检测肝功能生化指标,并进行小鼠主要脏器的采集、固定,对肝脏进行病理切片分析。结果:4种成分都可以使小鼠的肝脏系数明显的增加。大黄素组、鞣质组可以显著增加血清的ALT、AST、AKP、TP和AIB酶活力;且大黄素组可以显著降低IBIL的活性,鞣质组TBA酶的活性显著升高。光镜结果显示,鞣质对肝细胞的损害程度比较显著。结论:初步判定何首乌4种成分对肝脏都有一定程度的损害,但是实验结果表明,鞣质对肝脏的损害程度比较严重。 Objective:To study the effect of different components of Polygonum multiflorum on liver injury in mice. Methods:The four different components of Polygonum multiflorum emodin,chrysophanol,rhein,and tannins in accordance with 18.9 mg/kg,0.088 mg/kg,0.156 mg/kg,2290 mg/kg dosage were formulated with carboxymethyl cellulose sodium(CMCC). The mice were randomly divided into medication administration groups(4 groups). The stomach is administrated once a day(1 mL/50 g). The blank control group was given equal distilled water,for 4 weeks. At the end of the experiment,serum and liver tissue samples were collected to detect biochemical indicators of liver function,and mouse primary organs were collected and fixed,and liver sections were subjected to pathological section analysis. Results:All four components can be a significant increase in the mouse liver index. The emodin and tannin groups could significantly increased serum ALT,AST,AKP,TP and AIB enzyme activity;and emodin group could significantly reduced the activity of IBIL,and the activity of TBA enzyme in the enamel group was significantly increased. The results of light microscopy showed that tannins had more significant damage to hepatocytes. Conclusion:The preliminary determination of the four components of Polygonum multiflorumhas a certain degree of damage to the liver,but the experimental results show that the degree of damage of the liver to tannins is more serious.
