三种复合益生菌对菌群失调模型小鼠补偿生长和免疫功能的影响 被引量:6

摘要 为研究3种复合益生菌(Ⅰ号、Ⅱ号、Ⅲ号)对菌群失调模型小鼠补偿生长和免疫功能的影响,本实验以抗生素所致菌群失调模型小鼠为实验动物,对其造模后随机分为实验Ⅰ组(灌胃Ⅰ号菌)、实验Ⅱ组(灌胃Ⅱ号菌)、实验Ⅲ组(灌胃Ⅲ号菌)和不给予任何益生菌的正常组、模型组,实验期为30 d;自第20 d隔天1次,连续5次对小鼠肌内注射40 mg/kg氢化可的松(HY)进行免疫抑制。实验期结束后采用脏器系数测定法、单核-巨噬细胞吞噬功能测定法、绵羊红细胞(SRBC)诱导小鼠足趾增厚度法和半数溶血值(HC50)法,测定各组小鼠的平均周增重、脾脏指数、单核-巨噬细胞功能、细胞免疫功能和体液免疫功能。结果显示与模型组比较,持续补充Ⅰ号、Ⅱ号复合益生菌能有效促进处于免疫抑制状态菌群失调小鼠的生长(p<0.05);抵抗HY所致免疫抑制应激,促进小鼠脾脏发育(p<0.01),提高小鼠单核-巨噬细胞吞噬指数(p<0.05)、血清溶血素水平(p<0.01)、足趾增厚度(p<0.05)。以上结果表明,补充复合益生菌Ⅰ号和Ⅱ号对处于免疫抑制状态下菌群失调小鼠有明显缓解应激、补偿生长和提升机体免疫力的作用,为其在畜禽促生长和提升免疫力应用中提供实验依据。 To investigate the effects of three kinds of compound probiotics (Ⅰ ,Ⅱ and Ⅲ)on compensation growth and immune function in dysbacteriosis mice,the mice model of antibiotic-associated dysbacteriosis was utilized in this study.These mice were divided into four groups,Group Ⅰ (intragastric administration of bacterium Ⅰ ),Group Ⅱ (intragastric administration of bacterium Ⅱ ),Group Ⅲ (intragastric administration of bacterium Ⅲ),as well as the normal and model control groups without receiving any probiotics,with a trial period of 30days.The mice received five consecutive intramuscular injections with 40mg/kg hydrocortisone every other day since day 20to induce immunosuppression.Several index of the mice were examined to evaluate the effects of the probiotics on mice,including the mice average weekly weight gain,spleen index,the function of mononuclear macrophages,the cell-mediated immuity and humoral immunity,the organ index,mononuclear-macrophage phagocytic function, the approach of sheep erythrocyte (SRBC)induced toe thickness enhancement,and the half-hemolysis (HC50).The results showed that the mice in group Ⅰ and Ⅱ had quickly compensation growth and recover constitution (p<0.05),comparing with the mice in the model group;The immunosuppression of antagonistic hydrocortisone in the mice was able to promote spleen development (p<0.01),and improve mice mononuclear-macrophage phagocytic function (p<0.05),increase serum hemolysin level (p<0.01) and delay the type hypersensitivity (p<0.05).These results suggested that mice feeding with the compound probiotics (Ⅰ or Ⅱ )were able to significantly alleviate stress,compensate growth and enhance immunity of dysbacteriosis immunosuppression,comparing with the mice in model control group.This study provides a basis for facilitating the growth and enhancing the immunity in the livestock and poultry.
