水稻温敏叶绿素突变体叶片蛋白的双向电泳分析 被引量:15

Analysis of Leaf Protein of the Temperature-sensitive Chlorophyll Deficient Ri ce Mutants by Two-dimensional Electrophoresis

摘要 采用经改进的蛋白质双向电泳技术 ,分析了水稻温敏叶绿素突变体武金 4 B和 110 3S“斑马叶”性状表达过程中叶片蛋白质的变化。结果表明 :在该性状表达期 ,叶片蛋白质含量下降 ,特别是Rubis CO大、小亚基含量明显减少 ;在叶片的失绿部分蛋白 P1(MW5 6k D,p I6.7)发生特异缺失 ;在叶片的绿色部分该蛋白明显表达。复绿后 ,P1蛋白正常表达。在对照及未经变温刺激的武金 4 B叶片中该蛋白表达明显。由此推测 :P1蛋白可能是一种与叶绿素代谢过程密切相关的重要功能蛋白。此外 ,本文还就这种“斑马叶” The alteration of leaf protein of two temperature sensitive chlorophyll deficient rice mutants Wujin4B and 1103S was studied with two dimensional electrophoresis during the period when zebra leaf trait ap pea red. The results during the process of appearing zebra leaf showed that the protein content of leaves, especially the large subunit and small subunit o f RubisCO, decreased. A special polypeptide P 1 (MW56kD, p I 6.7) disappeare d in t he yellow area of leaves and existed still in the green area. P 1 polypeptide c an normally express in re greening areas of mutants leaves and in the leaves of t he control plants as well as in that of Wujin4B treated by unvaried temperature. It was suggested that P 1 protein could be an important functional protein rel ated with chlorophyll synthesis.
