摘要
目的 :用高效毛细管电泳法 ,以多索茶碱为内标物快速测定利巴韦林及中间体的含量。方法 :采用石英毛细管柱(未涂层 ) 5 7cm× 5 0 μm ,优化选择 2 5mmoL·L- 1 磷酸二氢钠 - 12 5mmoL·L- 1 硼砂 - 2 5mmoL·L- 1 SDS (pH 7 8)为电泳介质 ,操作电压 2 0kV ,柱温 2 3℃ ,测定波长 2 0 5nm。结果 :各物质间能达到很好分离 ,并具有良好的线性及回收率。结论 :方法准确且快速。
Objective:To establish a simple and rapid method for the assay of ribavirin and its 5 intermediate substances by high performance capillary electrophoresis (HPCE) using doxofylline as internal standard.Method:The experimental conditions were as follows,the electrophoretic medium was composed of 25 mmoL·L -1 NaH 2PO 4-12.5 mmoL·L -1 Na 2B 4O 7·10H 2O-25 mmoL·L -1 SDS(pH 7.8),uncoated capillary used was 57 cm×50 μm and detection with a UV monitor at 205 nm.The experimental voltage was 20 kV and the separation temperature was 23 ℃.Results:Each substance was well separated and had good linear relationship and recovery.Conclusion:The method is simple,rapid and specific.
