iASPP蛋白抗体的制备及iASPP在结肠癌组织中的表达及意义 被引量:2

摘要 目的研究iASPP多克隆抗体在结肠癌组织中的表达与结肠癌的组织分化程度、浸润深度、淋巴结转移的关系。方法根据生物信息学软件分析人iASPP蛋白的二级结构、亲水性、疏水性和抗原性等理化性质,经同源性检索,综合考虑抗体设计的其他因素,设计一段多肽,制备iASPP多克隆抗体。利用ELISA、Western印迹、免疫组织化学等方法来测定其效价和特异性。应用免疫组化(S-P)法检测30例结肠癌组织及15例正常结肠组织中的iASPP表达,判定其与结肠癌的组织分化程度、浸润深度、淋巴结转移等临床病理学特征的关系。结果制备了抗人iASPP多克隆抗体;利用ELISA,Western印迹和免疫组织化学染色的方法证实此抗体效价较高,特异性较强。iASPP在癌组织不同分化者iASPP阳性率不同。在浸润深度及有无淋巴结转移中的表达差异无显著性。结肠癌组织中iASPP的阳性表达率较正常结肠组织高(P<0.05)。结论 iASPP的表达异常与结肠癌的进展有关,其表达的失调在结肠癌的预后中发挥着重要作用。
