星点设计效应面法优化黄芩苷立方液晶纳米粒提取工艺的研究

摘要 目的采用星点设计效应面法优化黄芩苷立方液晶纳米粒(BCA—CLN)的处方。方法采用透射电镜(TEM)观察形貌;以粒径、包封率为评价指标,考察黄芩苷(BCA)加入量、单油酸甘油酯用量和F127用量对(BCA—CLN)性质的影响;根据归一值的最佳数学模型描绘效应面,选择最佳处方,并进行预测分析。结果黄芩苷立方液晶纳米粒呈类球形,三个影响因素和总评归一值之间存在定量关系。优选的处方范围为BCA用量为80—90mg,GMO用量为6.5~7g,F127用量为600—800mg。结论采用星点设计效应面法优化黄芩苷立方液晶纳米粒制备工艺,预测性良好。 Objective To optimize the formulation of Bacalin cubic liquid crystalline nanoparticles ( BCA - LCN) by a central composite design . Methods The optimal cubosome was characterized by transmission electron microscope (TEM) ; the effects of formulation variables (lipid concentration, F127 concentration, and drug load) on encapsulation efficiency and particle size of cu- bosomes were evaluated ; the response was described based on the optimal mathematical model of normalized value and further help to predict the formulation. Results The Bacalin cubosome was similar to round shape, there was a quantitative relationship between the three variables and normalized value , the optimized formulation was with 80 ~ 90 mg of BaIcalin,6.5 - 7 g of GMO, and 600 - 800 mg of F127. Conclusion Central composite design provide a good performance in the formulation optimization of Bacalin cu- bosomes.
