摘要
目的:建立赶黄草中槲皮素的含量测定方法;揭示乙醇浓度和提取温度与赶黄草中槲皮素含量之间的函数关系,优化出最佳赶黄草提取工艺。方法:采用高效液相色谱法测定槲皮素含量;以槲皮素含量为效应变量,以乙醇浓度和提取温度为考察因素,应用星点设计效应面法优选出最佳提取工艺。结果:槲皮素进样量在0.0541.08μg范围内与峰面积积分值呈良好线性关系(r=0.9999),平均回收率为98.54%,RSD-1.62%(n=6);赶黄草最佳提取工艺为乙醇浓度82.2%、提取温度84.5℃。结论:所建赶黄草中槲皮素含量测定方法的准确度、精密度和重现性均良好;所建数学模型能很好地预测槲皮素的提取效果,且准确度和重现性良好。
OBJECTIVE: To establish a method for determining the content of quercetin in Penthorum chinense and to study the functional relation between the extraction temperature and the ethanol concentration and the extraction efficiency of the quercetin in P.chinense so as to obtain the optimal extraction technology. METHODS: The content of quercetin in P.chinense was determined by HPLC. The optimal extracting technology was optimized by central composite design response surface methodology with the content of quercetin as variable and with ethanol concentration and extracting temperature as factors. RESULTS: A good linearity was obtained from 0. 054--1.08 μg( r = 0. 999 9) for quercetin. The average recovery was 98.54% (RSD - 1.62%, n = 6). The optimal technical parameters were as follows: the ethanol concentration was 82.2% and the extraction temperature was 84.5 ℃ .CONCLUSION: This established method is accurate, precise and reproducible; the extracting efficacy can be accurately and reproducibly predicted by the established mathematical model.
