摘要
杏鲍菇胞壁粗多糖经过羧甲基纤维素(CMC)离子交换柱、Sephadex G-75凝胶柱分离纯化得到一色谱纯多糖(PEWP)。采用高效液相色谱、红外光谱及化学分析等方法对PEWP的纯度、分子量及基本结构进行研究。结果表明:PEWP分子量为41 209 u,糖基组成为葡萄糖;红外光谱分析结果表明,PEWP具有多糖的特征吸收峰和典型的β-糖苷键构型;高碘酸氧化及Smith降解分析结果表明,PEWP中1→4糖苷键残基比例为83.3%,1→6糖苷键残基比例为12.8%,1→3糖苷键残基比例为2.4%,1→2糖苷键残基比例为1.5%。
Crude glucan from Pleurotus eryngii cell wall was subjected to CMC ion exchange chromatography and Sephadex G-75 gel column chromatography. Then a chromatographically pure gulcan (PEWP) was successfully isolated. The purity, molecular weight and structure of PEWP were measured through the high performance liquid chromatography (HPLC), infrared spectroscopy (IR) and chemistry analysis in the preliminary study. The results showed that the molecular weight of PEWP was 41 209 u and the monosaccharide unit was identified as glucose. IR analysis illustrated that PEWP had an absorption peak of {3-glucosidic bond, which belonged to the characteristic peak of polysaccharide. Periodate oxidation and smith degradation analysis showed that PEWP had 1→4 glucosidic bond (83.3%), 1→6 glucosidic bond (12.8%), 1→3 glucosidic bond (2.4%) and 1-.2 glucosidic bond (1.5%).
