摘要
建立新疆阿魏中不同部位阿魏酸的高效液相色谱含量测定方法。采用PLATISIL ODS (250 mm×4.6mm,5μm)色谱柱,甲醇-0.1%冰醋酸水溶液(35:65)为流动相,流速1.0mL/min,检测波长320nm,柱温30℃。测得阿魏酸进样量在0.440-5.396μg/mL(r=0.9997)范围内与峰面积有良好的线性关系,平均回收率达97.8%,RSD为1.79%(n=9)。新疆阿魏胶、根、茎和叶中的阿魏酸含量分别为0.2600、0.0362、0.0225和0.0275mg/g,胶中阿魏酸的含量远高于根、茎和叶。该方法简便、准确、重复性好,可用于新疆阿魏中不同部位阿魏酸含量的测定。
The objective of this study was to establish a HPLC method for the determination of ferulie acid in different parts of Ferula sinkiangensis. The HPLC method was performed on a PLATISIL ODS column (250 mm×4.6mm,5μm) with mobile phases consisted of methanol-0.1% glacial acetic acid in H2O (35: 65). The flow rate was 1.0 mL/ rain ,column temperature was 30℃ and the UV detection wavelength was set at 320 nm. Good linearity was obtained within the range of 0. 440-5. 396μm/mL ( r = 0.9997 ). The average recovery rate was 97.8% with RSD of 1.79% ( n =9). The concentration of ferulie acid in Fendic gum,root,stem and leaf of F. sinkiangengensis were 0. 2600,0. 0362, 0. 0225 and 0.0275 mg/g, respectively; the concentration of ferulic acid in Ferulic gum was much higher than that in root, stem and leaf. The HPLC method developed in this study was simple, accurate and reliable and thus was suitable for the determination of ferulie acid in different parts of F. sinkiangensis.
