超声对大鼠牙骨质修复过程中TGF-β_1表达和OPG/RANKL表达比值的影响 被引量:4

Effects of ultrasound on the expressions of TGF-β_1 and OPG/RANKL ratio during cementum reconstruction process in rat

摘要 目的:研究超声在大鼠牙骨质修复过程中对转化生长因子-β1(TGF-β1)表达和骨保护蛋白、核因子-KB受体活化因子配体(OPG/RANKL)表达比值的影响。方法:选择32只健康雄性Wistar大鼠建立正畸性牙根吸收动物模型,随机分为4组,每组8只,分别为空白对照组,单纯加力组,自然修复组,超声修复组,其中用频率1.5MHz,强度100MW/cm2的治疗超声对超声修复组的实验牙在2周的修复期进行每天15min治疗超声照射。2周治疗结束后,取上颌第一磨牙及其牙周组织,制作以上颌第一磨牙为中心的,近远中方向的组织切片,行TGF-β1,OPG,RANKL,免疫组化染色,光镜观察并作免疫组化光密度分析,对实验结果进行单因素方差分析。结果:超声修复组TGF-β1的光密度值(0.36228±0.02833),明显高于自然修复组(0.24576±0.03183),差异有统计学意义。超声修复组OPG/RANKL光密度值比值(1.1126±0.096),也明显高于自然修复组(0.7835±0.067)差异有统计学意义。结论:超声通过上调TGF-β1表达,改变OPG/RANKL的比值,促进牙骨质的修复重建。 Objective: To study the effect of ultrasound on the expressions of TGF-β1 and OPG/RANKL in the rat cementum reconstruction process.Methods: 32 healthy male Wistar rats were established as animal models of orthodontic root resorption,and randomly divided into four groups(the control group,experimental group,natural recovery group,ultrasound repair group) with 8 in each group.The ultrasound repair group were given 1.5MHz 100MW/cm2 ultrasound irradiation for 15min per day in the two weeks of repair period.Upper first molar-centered mesial-distal tissue slices were generated from the upper first molars and peridentium of the rats from each group.Specimen slices were analyzed with hematoxylin-eosin staining,OPG,RANKL,TGF-β1 immunohistochemistry,and optical microscopy.Analyses of densitometry,and one-way analysis of variance were performed.Results: expressions of TGF-β1 densitometry of ultrasound repair group(0.36228 ±0.02833)were significantly higher than natural repair group(0.24576 ±0.03183).expressions of OPG/RANKL densitometry ratios of ultrasound repair group(1.1126±0.096) were significantly higher than natural repair group(0.7835± 0.067).Conclusion: ultrasound promoted the repair and the reconstruction of the cementum by increasing the expressions of TGF-β1 and OPG/RANKL ratios.
