摘要
目的 :研究左旋精氨酸与氧自由基间的相互作用。方法 :14只离体大白鼠心脏模型 ,分别予含 5 %左旋精氨酸高钾停跳液 (实验组 )或单纯高钾停跳液 (对照组 ) ,使心脏停博 6 0分钟 ,再以KHBB缓冲液复灌 30分钟。观察复灌后鼠心的冠脉流量、心搏率和左室内压 ,并测定心肌匀浆内乳酸 (LA)、丙二醛 (MDA)、黄嘌呤氧化酶 (XOD)和超氧化物歧化酶 (SOD)含量。结果 :复灌时实验组冠脉流量明显多于对照组 ,鼠心搏率和节律恢复也早。实验组复灌时左室内压无显著变化 ,而对照组鼠心左室内压在收缩相和舒张相均明显上升 (36 %和 73 % )。复灌 30分钟时 ,对照组心肌匀浆LA ,MDA和XOD均比实验组分别高 2 0 % (P <0 0 5 )、2 6 % (P <0 0 1)和 36 % (P <0 0 0 1) ,而SOD仅为实验组的 87% (P <0 0 0 1)。结论 :左旋精氨酸可通过促进NO生成与释放 ,减少氧自由基的生成 ,进一步减少ONOO 的产生 ,从而减轻自由基对心肌细胞的损伤 ,达到心肌保护作用。
Objective:To study the interaction of L arginine and oxygen radicals.Methods:Fourteen Sprague Dawley rat hearts were randomly divided into two groups:control group(crystal cardioplegia,C group)and test group (ctystal cardioplegia+L arginine,T group).Before cardiac arrest and at 10,20 and 30 min after reperfusion,coronary blood flow(CBF),heart rates(HR) and left ventricular pressure(LVP) were observed.After reperfusion for 30 min,rat hearts were homogenized and the concentrations of lactic acid(LA),malonylaldehyde(MDA),superoxide dismutase (SOD),xanthine oxidase(XOD) in homogenate were measured.Results:After reperfusion CBF in T group was significantly higher than that in C group.No significant changes were found in LVP of T group after reperfusion,but the left ventricular systolic and diastolic pressures in C group increased significantly by 36% and 73% respectively after reperfusion.The concentrations of LA,MDA and XOD in C group were significantly higher than those in T group,but the concentration of SOD were lower.Conclusion:L arginine can reduce the generation of oxygen radicals and protect effectively myocardium from oxygen radicals during ischemia/reperfusion.
