摘要
目的观察罗格列酮(Ros)对高糖刺激下新生大鼠心脏成纤维细胞Ⅰ型前胶原表达的调节。方法 30 mmol/L高糖(HG)孵育新生大鼠心脏成纤维细胞,其中3组分别加入终浓度为20、30、50μmol/L的Ros干预12 h以检验不同浓度Ros对Ⅰ型前胶原表达的影响;另有3组均加入30μmol/L的Ros分别干预18、24、36 h以检验不同干预时间对Ⅰ型前胶原表达的影响;另设HG模型组(仅HG干预)及空白对照组[0.1%胎牛血清DMEM培养基+常规血糖浓度(NG)干预]各1组。应用Real-time PCR法测定心肌成纤维细胞Ⅰ型前胶原mRNA表达;Western blot检测Ⅰ型前胶原蛋白质的表达。结果 HG模型组Ⅰ型前胶原mRNA[(109±11)%]和蛋白(1.57±0.02)的表达与空白对照组[(40±3)%、(0.82±0.04)]比较显著升高,差异有统计学意义(P<0.05)。浓度依赖性实验结果显示,HG+10μmol/L Ros组Ⅰ型前胶原mRNA[(91±9)%]和蛋白(1.35±0.03)的表达低于HG模型组;HG+30μmol/L Ros组Ⅰ型前胶原mRNA[(62±5)%]和蛋白(1.08±0.04)的表达低于HG+10μmol/L Ros组;HG+50μmol/L Ros组Ⅰ型前胶原mRNA[(51±4)%]和蛋白(0.93±0.02)的表达低于HG+30μmol/L Ros组,差异均有统计学意义(P<0.05)。时间依赖结果显示,HG+30μmol/L Ros 18 h组Ⅰ型前胶原mRNA[(91±10)%]和蛋白(1.31±0.07)的表达低于HG模型组[(109±11)%、(1.57±0.02)];HG+30μmol/L Ros 24 h组Ⅰ型前胶原mRNA[(67±6)%]和蛋白(1.01±0.04)的表达低于HG+30μmol/L Ros 18 h组;HG+30μmol/L Ros 36 h组Ⅰ型前胶原mRNA[(42±4)%]和蛋白(0.87±0.01)的表达低于HG+30μmol/L Ros 24 h组,差异均有统计学意义(P<0.05)。结论 Ros可呈浓度和时间依赖性地下调高糖刺激下新生大鼠心肌成纤维细胞Ⅰ型前胶原的表达,可能在抑制心肌纤维化的过程中发挥了作用。
Objective To observe the regulation of Rosiglitazone (Ros) on the precollagen I expression of high glu- cose-induced neonatal rat cardiac fibroblast. Methods Neonatal rat cardiac fibroblast was incubated by 30 mmo]/L HG. Ros with the concentration of 20, 30, 50 μ mol/L were used to intervene for 12 hours, in order to examine the in- fluence of Ros on the precollagen I expression in different concentrations. Other 3 groups were given 30 μmo]/L Ros intervention for 18, 24, 36 hours, in order to examine the influence of Ros on the precollagen I expression in different times. Then a HG model group (only for HG) and a blank control group (0.1% fetal bovine serum DMEM medium+NG intervention) were settled. Real-time PCR was used to detect the precollagen I -mRNA expression of neonatal rat car- diac fibroblast. Western blot was used to detect the precollagen I -protein expression of neonatal rat cardiac fibroblast. Results Expression of preeollagen I mRNMprotein in HG model group [(109±11)%, (1.57±0.02)] were higher than those in blank control group [(40±3)%, (0.82±0.04)], the differences were statistically significant (P 〈 0.05). Concentration dependence experimental results showed that expression of precollagen I mRNMprotein in HG+10 μmol/L Ros group [(91±9)%, (1.35±0.03)] were lower than those in HG model group,
