摘要
目的运用脱氢表雄酮诱导SD幼年大鼠多囊卵巢综合征(PCOS)模型,并在药物注射剂量上对文献报道方法进行改良,研究模型的卵巢形态学和性激素等改变。方法采用23日龄SD雌大鼠,造模组每日皮下注射浓度为30mg/mL的脱氢表雄酮,注射量为0.2mL/100g体重,连续注射20d后。禁食于第21天测空腹血糖,处死测激素FSH、LH、T。结果造模组体重显著高于对照组(P<0.05)。造模组卵巢呈多囊样改变、闭锁卵泡增多,闭锁卵泡直径明显大于对照组。造模组血清T、FSH、空腹血糖水平明显高于对照组(P<0.05)。结论脱氢表雄酮诱导的多囊卵巢综合征模型的卵巢病理改变和激素改变与PCOS患者相似,是研究多囊卵巢综合征比较理想的模型。
Objective To induce the model of polycystic ovary syndrome in SD rats (PCOS) with the dehydroepiandrosterone, modify the reported drug infection does and study the morphological and sex hormone changes of ovary in the model. Methods The 23-day-old SD female rats were selected.The modeling group received continuous subcutaneous injection of 30mg/mL of dehydroepiandrosterone daily at the dose of 0.2 mL/100 g weight for 20 days,after which the rats fasted and received fasting blood glucose test on the 21st day,and after the rats were executed,FSH,LH and T hormones were measures. Results The modeling group had significantly higher weight than the control group (P 〈 0.05).The ovaries of the modeling group manifested polycystic changes and increased atretic follicles,of which the diameters were significantly larger than those of the control group.The modeling group was significantly higher than the control group in the serum T,FSH and fasting blood glucose levels (P 〈 0.05). Conclusion The pathological changes and hormone changes of ovary in the model of dehydroepiandrosterone-induced polycystic ovary syndrome in SD rats are similar to those of patients with PCOS, which is an ideal model of studying polycystic ovary syndrome.
