摘要
目的:比较两种多囊卵巢造模方法(雄激素造模方法和芳香酶抑制剂造模法)的实验差异。方法:通过监测2组造模大鼠动情周期、体质量、血清激素水平、卵巢大体解剖及光镜形态,比较两种造模方法各项指标的变化差异。结果:⑴DHEA组阴道脱落细胞为角质细胞;来曲唑组为小而圆的有核细胞。⑵体质量:DHEA组体质量增加幅度更显著。⑶血清激素水平:DHEA组血清T、P浓度增加更明显;DHEA组血清E2浓度升高而来曲唑组降低。⑷卵巢组织学:来曲唑组卵巢包膜增厚更明显,卵巢体积大;光镜下来曲唑组囊状扩张的卵泡沿包膜下排列,大小基本一致;DHEA组囊状扩张的卵泡占据卵巢髓质,体腔扩张明显。结论:脱氢表雄酮造模法更适合用来研究多囊卵巢综合征在体质量和血清激素水平方面的改变;来曲唑造模法则在卵巢组织学改变方面更突出。
Objective To analyze the experimental difference between two methods of polycystic ovary syndrome model making(dehydroepiandrosterone method and letrozole method)in rats.Methods The difference of indexes between two model groups were compared by monitoring the estrous cycle,weight,serum hormone level,general anatomy and light microscope morphology of ovary.Results ⑴The anovulia cells were keratinocyte in dehydroepiandrosterone group(DHEA group),the anovulia cells were nucleated cells in letrozole group;⑵the weight of rats in DHEA group were obviously increased than that of letrozole group;⑶the serum levels of testosterone and progesterone in DHEA group were increased obviously than those of letrozole group;the estradiol level was increased in DHEA group but decreased in letrozole group;⑷the thickness of ovarian capsule and volume of ovarium were obviously increased,the similar size of capsular dilatate folliculus were arranged along the ovarian capsule under light microscope in letrozole group.The capsular dilatate folliculus were occupied the ovarian medulla and the volume of ovarium were obviously dilated in DHEA group.Conclusion DHEA-induced model may be more suitable for evaluating the weight and serum hormone level changes,while the letrozole-induced model may be more prominent in observing the changes of ovarian histology in rats with polycystic ovary syndrome.
