摘要
目的观察羊栖菜多糖(Sargassum fusiforme polysaccharide,SFPS)对糖尿病脑缺血大鼠脑组织细胞凋亡的影响。方法通过HE染色观察SFPS对糖尿病大鼠脑组织形态学改变。通过TUNEL的方法检测脑组织细胞凋亡,并用RT-PCR、免疫组化的方法检测survivin基因mRNA和蛋白的表达的影响。结果SFPS可以明显改善糖尿病脑缺血大鼠脑组织病理形态改变。与模型组相比,SFPS组大鼠脑组织凋亡细胞显著减少,survivin基因的表达明显增加。结论SFPS具有保护糖尿病脑缺血损伤的作用,其作用机制可能与调控survivin基因的表达有关。
Objective To observe the effect of Sargassurn fusiforme polysaccharide(SFPS)on apoptosis of cerebrum tissue after cerebral ischemia injury in diabetic rats. Methods HE was used to stain hippocam- pal slices. Apoptosis was measured in cerebrum tissue with TUNEL method. Protein expression of Survivin was detected with immunochistochemistry and RT- PCR was used to measure rnRNA experes- sion. Results SFPS could improve the cerebrum diabetic model group, the number of apoptotic pathologic changes of diabetic rats. With comparison to cells in cerebrum tissue decreased and Survivin expres- sion increased in treated group. Conclusions SFPS could relieve brain damage induced by cerebra ische mia injury in diabetic rats, which may be related to regulating Survivin expression.
