调衡方对Lewis肺癌细胞增殖及JAK-STAT信号通路的影响 被引量:5

Effect of Tiaoheng Prescription on Proliferation and Signaling Transduction of JAK-STAT in Lewis Lung Cancer Cell Line

摘要 目的:观察调衡方对Lewis肺癌细胞增殖凋亡及JAK-STAT通路的影响。方法:实验动物分为4组,灌服给药,制备调衡方小、中、大剂量含药血清,正常血清。又将细胞分为5组,即正常血清组(不含药血清),小、中、大剂量含药血清组,中剂量含药血清+AG490(JAK-STAT信号通路特异性抑制剂)干预组,分别孵育Lewis肺癌细胞株24 h后,MTT法检测对Lewis肺癌细胞增殖的影响,吖啶橙染色观察Lewis肺癌细胞凋亡状况,RT-PCR法检测其对Lewis肺癌细胞Stat3、Stat5 mRNA的影响。结果:Lewis肺癌细胞经调衡方中、大剂量含药血清作用后,与对照组比较,细胞内Stat3含量降低至0.622±0.004、0.62±0.007(P<0.01),Stat5表达分别降低至0.700±0.034、0.649±0.033(P<0.01)。结论:调衡方含药血清能显著抑制Lewis肺癌细胞增殖,促进其凋亡,其机制可能与下调stat3、stat5的基因表达,抑制细胞信号转导通路有关。 Ohjective: To study the effect of Tiaoheng Prescription on proliferation and apoptosis and JAK-STAT signaling transduction pathway in Lewis lung cancer cell line. Methods : Experimental animals were divided into four groups: high-dose Tiaoheng Prescription drug-containing serum, middle-dose Tiaoheng Prescription drug-containing serum, small-dose Tiaoheng Prescription drug-containing serum and normal serum groups and drug-containing serum groups were made by administrating Tiaoheng Prescription. Furthermore, Lewis lung cancer cells were divided into five groups: high-dose drug-containing serum group, middle-dose drug-containing serum group, small-dose drug-containing serum group, middle-dose drug-containing serum + AG490 intervention group, normal serum group. After Lewis hmg cancer cells were incubated for 24 h, the inhibitory effect on Lewis lung cancer cells proliferation was detected by MTT; acridine orange fluorescence labeling was applied to measure the apotosis ratios of Lewis lung cancer cells ; the expressions of Stat3 and Stats mRNA in Lewis hmg cancer cell lines were assessed with RT-PCR ; the intracellular Star3 and Stats contents, expression levels of Star3 and Stat5 in the cells, and the conlents of Stat3 and Stats in the cells were detected. Results : Compared to normal serum group, Tiaoheng Prescription middle and high dose drug-containing serum groups could significantly decrease contents of Star5 to 0.622 ±0.004,0.62± 0.007 ( P〈0.01 ) and the expressions of Star5 to 0.70± 0.039,0.649±0.033 ( P〈0.05 ). Conclusion : Tiaoheng Prescription can inhibit the proliferation and induce the apoptosis of Lewis hmg cancer cells significantly. The mechanism might be related to down-regulating stat3 and star5 mRNA and inhibiting the signaling transduction pathway.
