摘要
目的:探讨阿魏酸通过调控JAK2/STAT6免疫信号通路抑制肺癌转移。方法:以A549细胞为研究对象,给药不同浓度阿魏酸,CCK-8检测A549细胞增殖变化;细胞划痕实验检测阿魏酸处理后A549细胞迁移能力变化;细胞侵袭实验检测阿魏酸处理后A549细胞侵袭能力变化;ELISA检测免疫因子IL-4、血小板衍生因子(PDGF)和粒细胞-巨噬细胞集落刺激因子(GMCSF)的表达;qRT-PCR和Western blot检测环氧化酶2(COX-2)、生存素(Survivin)、X连锁凋亡抑制蛋白(XIAP)和p53的mRNA和蛋白表达;Western blot检测阿魏酸处理A549细胞后P-JAK2和P-STAT6的变化。结果:CCK-8结果显示阿魏酸显著抑制A549细胞增殖;细胞划痕实验表明阿魏酸有效抑制A549细胞迁移;细胞侵袭实验显示阿魏酸能显著抑制A549细胞侵袭;ELISA结果显示阿魏酸有效降低免疫因子IL-4、PDGF和GM-CSF的表达;Western blot结果显示阿魏酸显著降低JAK2/STAT6的磷酸化及其下游凋亡蛋白的表达。结论:阿魏酸通过抑制JAK2/STAT6免疫信号通路从而抑制肺癌细胞增殖和转移。
Objective:To study ferulic acid inhibits lung cancer metastasis by regulating JAK2/STAT6 immune signaling pathway.Methods:Taking A549 cells as the research object,ferulic acid was administered at different concentrations,CCK-8 was used to detect the proliferation of A549 cells.Cell scratches experiments were used to detect changes in A549 cell migration capacity after ferulic acid treatment;cell invasion experiments were used to detect ferulic changes in invasion ability of A549 cells after acid treatment;ELISA detected the expression of immune factors IL-4,PDGF and GM-CSF;qRT-PCR and Western blot detected mRNA and protein expression of cyclooxygenase 2(COX-2),survivin(Survivin),X-linked apoptosis inhibitor protein(XIAP)and p53;Western blot detected the changes in P-JAK2 and P-STAT6 after ferulic acid treatment of A549 cells.Results:CCK-8 results showed that ferulic acid significantly inhibited the proliferation of A549 cells.Cell scratch experiments showed that ferulic acid effectively inhibited the migration of A549 cells.Cell invasion experiments showed that ferulic acid significantly inhibited the invasion of A549 cells.ELISA results showed that ferulic acid effectively reduced the expression of immune factors IL-4,PDGF and GM-CSF.Western blot results showed that ferulic acid significantly reduced the phosphorylation of JAK2/STAT6.Conclusion:Ferulic acid inhibits lung cancer cell proliferation and metastasis by inhibiting JAK2/STAT6 immune signaling pathway.
