巴戟天醇提物对离体大鼠心肌缺血再灌注损伤后白介素-1β、肿瘤坏死因子-α及心肌细胞凋亡的影响 被引量:12

摘要 目的探讨巴戟天醇提物对心肌缺血再灌注损伤(IRI)后白介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)水平以及心肌细胞凋亡的影响。方法取健康雄性Wistar大鼠80只,随机分为空白对照组、IRI组、巴戟天高剂量组和巴戟天低剂量组四组,每组各20只。利用Langendorff灌流技术制备心肌IRI模型,两个巴戟天用药组分别在预灌时给予相应浓度的药物。灌流结束后,各组取10只大鼠左心室组织,采用ELISA法检测其IL-1β、TNF-α的表达;取另外10只大鼠左心室组织,采用TUNEL法检测其心肌细胞凋亡情况。结果与IRI组相比,巴戟天高剂量组和巴戟天低剂量组IL-1β、TNF-α表达降低(P<0.05),凋亡指数降低(P<0.05)。结论巴戟天醇提物能够减轻心肌IRI后心肌细胞凋亡,这可能与其能够降低心肌组织中的IL-1β、TNF-α表达有关。 Objective To investigate the influence of radix morindae officinalis ethanolic extracts on levels of interleukin1β(IL-1β),tumor necrosis factor-α(TNF-α) and myocardial cell apoptosis in rats with myocardial ischemia-reperfusion injury(IRI).Methods 80 healthy male Wistar rats were randomly divided into 4 groups:control group,IRI group,high dose radix morindae officinalis group and low dose radix morindae officinalis group,each group had 20 rats.Myocardial IRI model of rat was established with Langendorff method,and all the medication groups were given with corresponding concentration drugs before perfusion.After perfusion,the expressions of IL-1 β,TNF-α in myocardium were determined by ELISA method in 10 rats of each group;the myocardial cell apoptosis in myocardium was determined by TUNEL method in the other 10 rats of each group.Results Compared with the IRI group,the expressions of IL-1β,TNF-α decreased significantly in the high dose radix morindae officinalis group and the low dose radix morindae officinalis group(P0.05),and the myocardial cell apoptotic indexes decreased significantly too(P0.05).Conclusion Radix morindae officinalis ethanolic extracts can decrease the myocardial cell apoptosis in rats with myocardial IRI,this may be related to the decreases of IL-1β and TNF-α content in myocardium.
